These findings demonstrate markedly improved PRS accuracy for BC risk prediction in African-ancestry women. Using these PRS models for screening will help promote more equitable cancer prevention efforts.
SERENA-3 demonstrates that camizestrant 75 mg once daily (Phase III dose) is well-tolerated and achieves maximal reduction in ER through known mechanisms, that is, antagonism and degradation, by 5-7 days, and proliferation suppression determined by Ki67 expression, by 12-15 days. These data support camizestrant 75 mg once daily as the preferred dose for ongoing clinical development and highlight the importance of presurgical WOO studies in guiding dose selection.
This extraordinarily rare case of axillary skip metastasis limited to the interpectoral node emphasizes the potential for false-negative SN biopsies. Careful review of preoperative images, particularly MRI images, is crucial to avoid understaging. Awareness of interpectoral node involvement may help guide appropriate treatment strategies for selected patients.
Spatial and coculture analyses of these tumors demonstrated that the CD8+ T cell-associated chemokine CXCL11 drove estrogen-independent tumor growth. These findings identify an immune-mediated mechanism of endocrine resistance in breast cancer and identify CXCL11 as a potential biomarker and therapeutic vulnerability.

The ESGO-ESTRO-ESP 2025 classification re-assigns approximately one-tenth of patients, with a meaningful impact on adjuvant therapy. The rarity of estrogen receptor negativity limits its role in no-specific-molecular-profile stratification. Variation in lymph node involvement across molecular tumor categories highlights opportunities for individualized surgical staging.
An ALC ≥ 1000/μL at ERI completion was associated with an improved post-ERI OS (OS2). It is suggested that not only the factors at the initiation of ERI, but also the immunological status at the end of ERI, may have prognostic value after ERI.
Many of deregulated genes were directly bound by all three proteins, suggesting a coordinated transcriptional regulation, revealing a critical axis involving ERα, DOT1L, menin and PVT1. Targeting this RNA-dependent chromatin associated regulatory complex could offer novel therapeutic strategies for BC treatment.
