Differential long-term tamoxifen therapy benefit by menopausal status in breast cancer patients: secondary analysis of a controlled randomized clinical trial. (PubMed, J Natl Cancer Inst)
Our study suggests differential tamoxifen benefit by menopausal status. Improved long-term endocrine therapy prediction in premenopausal patients is needed and could involve molecular markers because standard tumor characteristics cannot predict benefit beyond 10 years.

Association between long-term ambient air pollution exposure and the incidence of breast cancer: A meta-analysis based on updated evidence. (PubMed, Ecotoxicol Environ Saf)
This study is the first to document a significant association between long-term exposure to ambient PM2.5 and breast cancer incidence through meta-analysis. Air pollution has a pronounced impact on postmenopausal breast cancer, and the strength of the association between specific air pollutants and breast cancer incidence varies across regions. These findings suggest that long-term exposure to NO2 and PM2.5 may increase the incidence of breast cancer.

Early Breast Cancer Treated with Adjuvant S-1 plus Endocrine Therapy-A Report of Two Cases (PubMed, Gan To Kagaku Ryoho)
We judged that both patients were at intermediate risk of recurrence and administered S-1 in addition to endocrine therapy. Even in cases without axillary lymph node metastasis, this combination of S-1 with endocrine therapy was considered to be a useful treatment option if the criteria of the POTENT trial were met.

Long-Term Response to CDK4/6 Inhibitor for Multiple Metastases of Breast Cancer (PubMed, Gan To Kagaku Ryoho)
Surgery was not possible due to comorbidities, so fulvestrant(500 mg/month)+denosumab(120 mg/month)was started. Furthermore, as the tumor markers were elevated, abemaciclib(300 mg/day)was added, which resulted in a decrease in the tumor markers...During the course of treatment, the tumor markers rose again, so the dose was increased to 250 mg/day, which resulted in a decrease in the tumor markers and good tolerability. At present, 36 months after the start of treatment, long SD has continued, no adverse events of grade 3 or higher have been observed, and the drug has been well tolerated.

Metastatic disease in patients with early breast cancer and a low Oncotype DX® Recurrence Score: A review of clinicopathological features (SABCS 2024)
57.14% (4/7) received adjuvant Letrozole for a minimum of 5 years. The patient with a recurrence score of 24 received adjuvant chemotherapy and developed metastatic 4 years post diagnosis while on Tamoxifen...Genomic tests have resulted in a significant reduction in the number of patients receiving adjuvant chemotherapy. However, a more multifacted approach may be required to tailor treatment decisions incorporating genomic testing, clinicopathological features and individual characteristics with the aim of improving outcomes for patients with hormone receptor positive early breast cancer.

Prognostic Value of high-risk Oncotype Recurrence Score (RS) and MammaPrint (MP) assay in Premenopausal African American (AA) Patients with Hormone-Positive (HR+), Node-Negative Breast Cancer (BC): A Study of the National Cancer Database (NCDB). (SABCS 2024)
Our study shows that among high genomic risk, node-negative, premenopausal patients, the survival rates are numerically similar between Caucasians and African Americans (AAs) when chemotherapy was administered. However, when chemotherapy was omitted, there were numerical differences in the survival rates within the high-risk RS population. This trend was not observed with the MP cohort.

The 70-Gene Signature Assay vs. Traditional Clinical Risk Assessment: Impact on Adjuvant Chemotherapy Choices in Mexican Breast Cancer Patients (SABCS 2024)
The NPI value is calculated based on tumor size, number of lymph nodes and tumor grade (11), The PREDICT program algorithm was developed from data of the East Anglia Cancer Registry and Information Centre (ECRIC) on patients with primary breast cancer treated in 1999-2003 and provides 5- and 10-year survival estimates with and without adjuvant systemic therapy (hormonal therapy, chemotherapy and trastuzumab) (13)...Although limited by sample size and lack of follow-up, these findings emphasize the value of integrating genomic tools with conventional methods to enhance personalized treatment. Future large-scale studies with follow-up are needed to confirm these results and fully establish the clinical utility of the 70-Gene Signature Assay in Mexican patients.

Utility of 18F-fluoroestradiol over 18F-fluorodeoxyglucose positron emission tomography/computed tomography in the initial diagnosis of over 90 metastatic lesions in a patient with metastatic estrogen receptor-positive breast cancer. (PubMed, Proc (Bayl Univ Med Cent))

Reductions in recurrence in women with early breast cancer entering clinical trials between 1990 and 2009: a pooled analysis of 155 746 women in 151 trials. (PubMed, Lancet)
Most of the improvement in trial outcomes is explained by a greater proportion of women with lower-risk disease entering trials and improved adjuvant treatment. After adjustment, women diagnosed since 2000 have about a fifth lower rate of distant recurrence than the 1990s. Long-term risks of distant recurrence for oestrogen receptor-positive disease remain, but are about a tenth lower now than in our previous report.

Single-cell multiplex immunocytochemistry in cell block preparations of metastatic breast cancer confirms sensitivity of GATA-binding protein 3 over gross cystic disease fluid protein 15 and mammaglobin. (PubMed, Cancer Cytopathol)
For a single marker, GATA3 displayed the highest sensitivity. The addition of MMG for hormone receptor-positive breast cancers and GCDFP15 for hormone receptor-negative breast cancers further increased sensitivity. The low proportion of multimarker-positive cells suggested that the coexpression observed with traditional ICC is attributable to intratumoral heterogeneity, not genuine coexpression.
