80% of UMs harbor mutations in the Gαq family members GNAQ and GNA11....We exposed two GNAQ mutants, one GNA11 mutant, and two melanoma cell lines without either mutation to combinations of AEB071 and PD0325901 at doses ranging from 0 to 5μM and 0 to 0.5μM, respectively...We observed an improved growth inhibition of the combinations in all three melanoma cells with GNAQ/11 mutation (92-1, OMM1.3 and OMM-GN11) compared to treatments with either compound alone.
