EIF4A3-induced circEIF2S2 facilitates colorectal cancer growth, metastasis, and immune suppression via the miR-646/UHMK1 Axis. (PubMed, Int J Biol Macromol)
In vivo, circEIF2S2 depletion significantly inhibited tumor growth and liver metastasis in xenograft models. Collectively, these findings identify the EIF4A3-circEIF2S2-miR-646-UHMK1 axis as an important regulatory pathway involved in CRC progression and tumor-associated immunosuppression.

Moreover, ectopic BART miRNA expression in several EBV-negative BL cells induced transcriptional changes that may influence molecular signatures of EBV-associated BLs. Collectively, our findings reveal novel, functional interactions for BART miRNAs in lymphomas and provide insights into their roles in these B cell cancers.

However, some of these genes have a common role: APBB2, RUNX1T1, ZBTB7C, and ISX regulate transcription, while APBB2, BTG3, ZBTB7C, SZRD1 and LEF1 have a function in regulating proliferation, suggesting a role for these two pathways. While our results are specific for the lung cancer cell lines we selected for this work, our method of analysis can be applied to cell lines from other tissues and for which the required data is available.

The complementary experiments for cell growth and oxaliplatin resistance indicated the interdependent relationship between UHMK1 and STAT3. Thus, these collective findings uncovered a new UHMK1/STAT3 positive feedback regulatory loop contributing to CRC development and chemoresistance.
