SOD1 at the Crossroads: Co-Overexpression of Canonical Antioxidant Response and Noncanonical Hydrogen Sulfide Generation Pathways in Down Syndrome, With Immune Cell Implications. (PubMed, Res Sq)
Conversely, the increased expression of a thioredoxin-dependent cysteine catabolism pathway suggests noncanonical H 2 S overproduction in DS distinct from CBS . Our findings highlight the unexpected relationship between oxidative stress homeostasis and H 2 S overproduction in DS, extending beyond Hsa21 trisomy.

Curcumin alleviates ferroptosis in CCl₄-induced acute liver injury in vitro: involvement of TXNIP/NLRP3 inhibition and SLC7A11/GSH/GPX4 activation. (PubMed, Naunyn Schmiedebergs Arch Pharmacol)
It also reduced total superoxide dismutase (T-SOD), glutathione (GSH), thioredoxin-1 (Trx-1), glutathione peroxidase 4 (GPX4), SLC7A11, and mitochondrial membrane potential, leading to mitochondrial damage. CUR effectively reversed these changes by inhibiting oxidative stress and ferroptosis through the TXNIP/NLRP3 pathway and the SLC7A11/GSH/GPX4 axis.

METTL1-driven epitranscriptomic enhancement of TXNDC12 boosts c-Myc stability through USP5 in HNSCC. (PubMed, Exp Mol Med)
Modulating TXNDC12 levels demonstrates that its reduction curtails aggressive tumor phenotypes and cisplatin resistance, while its overexpression exacerbates these characteristics...Clinical validation underscores the importance of the METTL1-TXNDC12-c-Myc axis in HNSCC. Our findings reveal that METTL1-coupled epitranscriptomic upregulation of TXNDC12 in HNSCC enhances c-Myc signaling by promoting its USP5-mediated stability.

The expression characteristics of TXN in pan cancer and its impact on tumor immunity and prognosis (PubMed, Xi Bao Yu Fen Zi Mian Yi Xue Za Zhi)
Conclusion TXN may become a potential prognostic biomarker in pan cancer, providing strong theoretical basis for future tumor diagnosis and prognosis judgment. The retinoic acid-inducible gene-I (RIG-I)-like receptor signaling pathway, Toll-like receptor (TLR) signaling pathway, and nucleotide binding oligomerization domain (NOD)-like receptor signaling pathway may be crucial pathways through which TXN influences tumor immunity.

Thioredoxin: a key factor in cold tumor formation and a promising biomarker for immunotherapy resistance in NSCLC. (PubMed, Respir Res)
Furthermore, TXN knockdown enhanced T-cell-mediated cytotoxicity against these tumor cells, suggesting that TXN contributes to immune escape in NSCLC by promoting tumor cell proliferation and migration while inhibiting immune killing. Notably, TXN knockdown also upregulated CD40 expression, indicating that TXN may regulate immune escape in lung cancer through CD40 modulation.

Integrated Bioinformatic Analyses Reveal Thioredoxin as a Putative Marker of Cancer Stem Cells and Prognosis in Prostate Cancer. (PubMed, Cancer Inform)
Despite the limited sample size in our study and the need for further in vitro and in vivo experiments to demonstrate whether TXN functionally regulates prostate CSCs, our findings suggest that TXN may serve as a novel therapeutic target against CSCs. Moreover, TXN expression in CSCs could be a useful marker for predicting the prognosis of PCa patients.

Hydrogel PCDME creates pancreatic cancer stem cells in OXPHOS metabolic state with TXNIP elevation. (PubMed, Biochem Biophys Res Commun)
In metabolic analysis, CSCs showed a shift of energy production from glycolysis to oxidative phosphorylation (OXPHOS). Furthermore, knockdown of TXNIP on PCDME gel using shRNAs decreased growth speed and in vivo tumorigenicity, suggesting that TXNIP may be involved in CSCs induction.

The Modulatory Effect of Selol (Se IV) on Pro-Inflammatory Pathways in RAW 264.7 Macrophages. (PubMed, Int J Mol Sci)
Our study also showed that as a result of LPS action in cells, the anaerobic glycolysis activity was increased, and incubation with Selol caused a partial reprogramming of Glc metabolism towards aerobic metabolism. This may indicate different pharmacological and molecular effects of Selol action in physiological and pathological conditions.

TXN promotes tumorigenesis by activating the ERK1/2 and ERK5 signaling pathways regulating c-Myc in non-small cell lung cancer. (PubMed, Cell Signal)
Our results indicated tumor promotion of TXN in NSCLC and TXN regulated c-Myc in the interest of tumorigenesis through ERK1/2 and ERK5 signaling pathways. Targeting TXN and blocking the ERK1/2 and ERK5 pathways could potentially offer new therapeutic strategies for NSCLC.
