TRPS1 Expression Across the Spectrum of Paget Disease and Morphologic Mimics. (PubMed, Appl Immunohistochem Mol Morphol)
Among EMPD cases, perivulvar (n=10) and periscrotal (n=5) cases were consistently positive, whereas perianal lesions (n=10) showed no/weak expression. In summary, TRPS1 demonstrates universal expression in mammary PD of all ER/HER2 subgroups and in Toker cells, and corroborates recent reports regarding the utility of TRPS1 in site-of-origin studies.

Identification of Resistance Genes in Breast Cancer Cells Treated with Fulvestrant and Ribociclib via Retroviral Screening and Integration Site Sequencing. (PubMed, Cells)
ER-positive ZR75.1 breast cancer cells transduced with retroviral vectors were treated with endocrine (tamoxifen, fulvestrant) or CDK4/6i monotherapies (abemaciclib, palbociclib, ribociclib) or a combination of fulvestrant and ribociclib. Twenty of these loci were also identified as candidates for resistance to other CDK4/6i and to fulvestrant and ribociclib combination therapy, including TRPS1 and TRIM24-genes that are involved in resistance to endocrine therapy but have not yet been associated with resistance to CDK4/6i. The identification of unique and shared resistance-associated loci highlights the complexity of resistance pathways.

The evolving role of TRPS1 in dermatopathology: insights from the past 4 years. (PubMed, J Pathol Transl Med)
In this review, we present the most comprehensive and up-to-date evaluation of the utility and limitations of TRPS1 immunohistochemistry in dermatopathology. Our aim is to deepen understanding of this emerging marker and provide practical guidance on its optimal integration with established immunohistochemical panels to enhance diagnostic accuracy in routine practice.

We propose that the initial IHC panel should include TRPS1, CK7 and CK20. In TRPS1-negative cases, additional immunostains should be performed: CDX2 and SATB2 for colonic; p63, GATA3 and uroplakin II/III for urothelial; and PSA and NKX3.1 for prostatic secondary EMPD.

Identification of an efficient immunohistochemical panel for precise differential diagnosis of sebaceous carcinoma of eyelid. (PubMed, Discov Oncol)
In addition, TRPS1 exhibited higher AUC values than GATA3 and adipophilin in differentiating primary and recurrent SCs, although the difference was not significant. Collectively, our findings indicate that the TRPS1/GATA3/adipophilin-based model demonstrated excellent discriminatory efficacy for eyelid malignancies (Kappa = 0.784, micro-average F1 = 0.882, AUC-ROC = 0.964 [95%CI:0.934-0.994]), achieving clinical-grade performance.

TRPS1::PLAG1 Fusion in a Primary Cutaneous Myoepithelial Carcinoma: A Case Report and Literature Review. (PubMed, J Cutan Pathol)
Here, we describe a case of occipital primary cutaneous myoepithelial carcinoma occurring in a 56-year-old patient. RNA sequencing was performed on the tumor which identified a TRPS1::PLAG1 fusion event which is to our knowledge, the first time this has been reported in the literature for a primary cutaneous myoepithelial carcinoma.
