Elevated co-expression of TIMM17A and NMT1 is associated with poor survival in non-small cell lung cancer. (PubMed, Sci Rep)
This highlights a novel NMT1/TIMM17A axis as a promising pathway for predicting patient prognosis. Further studies are warranted investigating both genes as targets for novel therapeutic strategies for KRAS mutant non-small cell lung carcinoma with STK11 and/or KEAP1 co-mutations.

HSF1 remodels mitochondrial biogenesis and function in cancer cells via TIMM17A. (PubMed, bioRxiv)
The HSF1- HSPD1/HSPE1-TIMM17A axis remodels the mitochondrial proteome to promote mitochondrial translation and energy production, thereby supporting robust cell proliferation. Our work reveals a mechanism by which mitochondria adjust protein uptake according to the folding capacity in the matrix by altering TIM complex composition.

MicroRNA-593-3p Inhibits Growth and Metastasis of Human Breast Cancer Cells by Regulating TIMM17A Expression. (PubMed, Biochem Genet)
Transwell assays further demonstrated that miR-593-3p reduced the invasive potential of MDA-MB-231 cells by regulating TIMM17A These findings indicate that miR-593-3p functions as a tumor suppressor by targeting TIMM17A, influencing both proliferative and invasive properties of breast cancer cells. The identification of this regulatory axis suggests that miR-593-3p could serve as a promising therapeutic target for breast cancer treatment.

Myricetin ameliorates the effects of hydrogen peroxide-induced oxidative stress in human mesenchymal stem cells: an ultrastructural and immunocytochemical study. (PubMed, Ultrastruct Pathol)
The results suggest that myricetin prevents oxidative stress-induced apoptosis and inflammation in the HUC-MSCs. Myricetin can be combined with HUC-MSCs in cell culture and considered as a supportive alternative treatment option.

Sulforaphane downregulates mitochondrial TIGAR via inhibiting mitochondrial transmembrane assembly and LONP1/CASP3 axis causing apoptosis. (PubMed, Biochem Biophys Res Commun)
Further, SFN downregulated mitoprotease LONP1 and decreased the binding of LONP1 to TIGAR; knockdown of LONP1 activated mitochondrial caspase-3 causing the cleavage of mitochondrial TIGAR; SFN-mediated the reduction of TIGAR decreased NADPH leading to ROS elevation and apoptosis in NSCLC. These studies will provide key targets for anti-NSCLC therapy.

OCIAD1 and prohibitins regulate the stability of the TIM23 protein translocase. (PubMed, Cell Rep)
The expression of OCIAD1 was in turn regulated by the status of the TIM23 complex. We postulate that OCIAD1 together with prohibitins constitute a regulatory axis that differentially regulates variants of human TIM23.

This study reveals the unexpected connection between protein myristoylation, the mitochondrial import machinery, and iron homeostasis. It also uncovers myristoylation inhibitors as novel inducers of parthanatos in cancer, and the novel axis N-myristoyltransferase-TIM17A as a potential therapeutic target in highly aggressive lung carcinomas.

Knockdown of TIMM17A inhibited the growth of LUAD cells. The potential of TIMM17A as a biomarker and therapeutic target for LUAD presents a promising pathway for improving patient diagnosis and treatment strategies.

Invasive Breast Cancer: miR-24-2 Targets Genes Associated with Survival and Sensitizes MDA-MB-231 Cells to Berberine. (PubMed, OMICS)
In all, this study reports on the novel target genes of miR-24-2 in invasive breast cancer/TNBC, and that miR-24-2 sensitizes MDA-MB-231 cells to berberine. These data lend evidence for the translational potentials of miR-24-2 for invasive breast cancer diagnostic and therapeutic innovation.
