This multi-omics network analysis reveals a shared molecular framework between IBD and CRC, with inflammation as a key driver of colorectal carcinogenesis.

Similar durable TFF1 repression was observed in primary gastric mucosoids following exposure to inflammatory mediators. Overall, our findings show that transient inflammatory signals cause durable gene silencing through epigenetic remodeling, revealing how chronic inflammation can reprogram epithelial cells and promote cancer, while suggesting strategies to reverse these effects.

Our results indicate that low BC TFF1 expression is associated with younger age and aggressive tumor features, on top of serving as a predictor of the basal-like subtype. High TFF1 protein expression in LN metastases may carry prognostic significance for younger patients.

The integration of AH liquid biopsy in RB management may aid prognosis prediction and optimize treatment strategies. However, further research is needed to validate the prognostic significance.

This study uncovers a previously unrecognized role for BPCs in UC pathogenesis, highlighting their spatial dysfunction and potential as a therapeutic target. The identified FABP1-led matrix offers new insights into epithelial dysregulation in UC and its progression to colorectal cancer, suggesting that restoring BPC function may provide novel treatment avenues.

BPA also increased cell numbers, and both effects were reversed by fulvestrant. Collectively, we identified BPTMC as a potent ERα agonist capable of eliciting transcriptional and mitogenic responses at low concentrations, raising concerns about its endocrine-disrupting and breast cancer-promoting effects.

Importantly, we identified TFF1 as the core gene, and the immunohistochemical (IHC) assay using the tissue microarray revealed that TFF1-positive patients had significantly better OS than their counterparts. We constructed a highly precise clinical prediction model based on SERS, demonstrating the impact of SE-associated genes on the prognosis of BC patients and their regulatory effects on somatic mutations, tumor immune microenvironment, and drug sensitivity in BC.

Our findings underscore the prognostic value of the identified genes and the immunosuppressive TME in TP53-mutant breast cancer. The identification of drug candidates with strong binding affinities to key proteins provides promising avenues for targeted therapy in this high-risk patient population.

This study provides a spatially resolved molecular map of IPMN progression, delineating key transcriptomic and immune signatures. These findings advance the understanding of IPMN biology and highlight potential biomarkers for risk stratification and therapeutic strategies.

Pre-processing with dual-feature filtering and feature extraction via lifted Euler characteristic transform enhanced data quality.
