This study evaluated the immune surveillance function in chronic myeloid leukemia (CML) through tumor antigen gene expression analysis, in vitro mixed lymphocyte cultures with peptides, and immunological analysis of patients after imatinib withdrawal to investigate factors affecting treatment-free remission (TFR)...A Cox model confirmed the prognostic value of immune markers. These results emphasize the critical role of the immune system in CML and indicate novel targets for sustaining TFR.

We also discuss the incorporation of CTA testing in lung cancer diagnostic workflows with focus on its analytical performance, preanalytical factors as well as comparative value vs existing markers. Finally, we outline the role of CTAs as companion diagnostics in immunotherapy and targeted treatment plans, and indicate the main challenges and research priorities towards the clinical translation of CTAs.

Truncated SPAG9 as a novel candidate gene for a new syndrome: Coarse facial features, albinism, cataract and developmental delay (CACD syndrome). (PubMed, Genet Mol Biol)
For the first time in the literature, biallelic SPAG9 gene variation was linked to multisystem-exhibiting features like coarse facial features, albinism, cataracts, skeletal abnormalities, and developmental delay. Thus, this data supports the notion that SPAG9 plays an important role in a multisystemic disorder in humans.

Synergistic Role of Amino Acids in Enhancing mTOR Activation Through Lysosome Positioning. (PubMed, bioRxiv)
In summary, amino acid-regulated lysosome positioning and mTOR signaling depend on distinct sets of amino acids. Combining lysosome-dispersing amino acids with mTOR-activating amino acids synergistically enhances mTOR activation, which may be particularly relevant in cancer cells.

Proteomic Characterization of Urinary Exosomes with Pancreatic Cancer by Phosphatidylserine Imprinted Polymer Enrichment and Mass Spectrometry Analysis. (PubMed, J Proteome Res)
Mass Spectrometry with nontargeted proteomics was combined to show differentially expressed proteins in exosomes between 5 pancreatic cancer patients and 5 healthy controls. The most significant changes in the proteomic profile in pancreatic cancer patients compared to healthy controls were the overexpression of SLC9A3R1, SPAG9, and ferritin light chain (FTL) These proteins may have an important role in diagnosis and prognostic assessment, supporting further scientific and clinical studies on pancreatic cancer.

The findings suggest that immune cell infiltration profiles play a significant role in the shared pathogenesis of smoking-related OP and COPD. The results could provide valuable insights for developing novel therapeutic strategies for managing these disorders, as well as shedding light on their pathogenesis.

SPAG9 Expression Predicts Good Prognosis in Patients with Clear-Cell Renal Cell Carcinoma: A Bioinformatics Analysis with Experimental Validation. (PubMed, Genes (Basel))
We further found that some genes might cooperate with SPAG9 to promote autophagy, and that these were highly expressed in the tumor stroma and could be represented by key genes. The SPAG9-based nomogram can help to estimate the long-term prognosis of ccRCC patients, indicating that SPAG9 is a potential prognostic marker for ccRCC.

An HLA-G/SPAG9/STAT3 axis promotes brain metastases. (PubMed, Proc Natl Acad Sci U S A)
Importantly, we show that premetastatic BMICs exhibit increased expression of HLA-G, which we further demonstrate functions in an HLA-G/SPAG9/STAT3 axis to promote the establishment of brain metastatic lesions. Our findings suggest that unraveling the molecular landscape of premetastatic BMICs allows for the identification of clinically relevant targets that can possibly inform the development of preventive and/or more efficacious BM therapies.
