The combined four-gene model demonstrated moderate discriminatory power, yielding an AUC of 0.76 (95 % CI 0.65-0.86, p < 0.0001) with balanced sensitivity and specificity of 69.8 % each at the 0.5 cutoff (overall accuracy 69.8 %). Taken together, RUSC1-AS1, SNHG17, PABPC1L and SNHG1 can be novel candidates for future diagnostic studies in CRC.

Pharmacological interventions targeting autophagy (Rapamycin) and Notch signaling (DAPT) further confirmed that SNHG1's effects on EMT are mediated via modulation of the autophagy-Notch1 interplay...Additionally, SNHG1 expression was significantly elevated in patients who progressed to low- or high-grade dysplasia, as confirmed by diagnostic endoscopic biopsies. Collectively, our study uncovers SNHG1 as a central molecular mediator linking acidic bile-induced EMT and autophagy regulation via the ULK1-Notch1 axis, highlighting its potential as a therapeutic target for preventing BE recurrence and progression to EAC.

Elevated SNHG1 promotes invasion and migration of Cd(II)-transformed cells through Sox2, Rac1, and Slug. (PubMed, Toxicol Appl Pharmacol)
Additionally, we observed increased expression of Slug, a key epithelial-mesenchymal transition (EMT) transcription factor, and decreased expression of its downstream target E-cadherin in Cd(II)-transformed cells. Collectively, these results demonstrate that elevated SNHG1 promotes the expression of Sox2, Rac1, and Slug, thereby driving the invasive and migratory behavior of Cd(II)-transformed cells.

Therefore, this review aims to summarise the current investigations on the pathogenesis of SNHG1 in HCC, highlighting its potential as a molecular marker for early prediction and prognostic assessment.
