Repositioning serotonergic antagonists as therapeutic agents in gastric cancer: induction of apoptosis, inhibition of cell migration, and cell cycle arrest in AGS cells. (PubMed, Cytotechnology)
Imipramine induced G1/S phase arrest, whereas tropisetron, ketanserin, and cyproheptadine increased the sub-G1 cell population. Gene expression analysis revealed decreased Bcl-2 and PCNA levels and increased Bax expression.These findings suggest the potential of tropisetron, imipramine, ketanserin, and cyproheptadine as repurposed therapeutic agents for gastric cancer.

Serotonin modulates lineage plasticity in neuroendocrine prostate cancer via epigenetic reprogramming. (PubMed, Cancer Discov)
Mechanistically, high level of intracellular serotonin promotes histone serotonylation at H3K4me3Q5, reconfiguring the H3K4me3 chromatin landscape and downstream gene expression, which drives induced NE differentiation and is associated with suppressed androgen receptor signaling. Pharmacological inhibition of 5-HT synthesis using the FDA-approved DDC inhibitor carbidopa significantly impairs tumor growth and prolongs survival in both genetically engineered and patient-derived xenograft models, highlighting histone serotonylation as a druggable vulnerability in NEPC.

Impact of pesticide diazinon on gastric cancer cell lines and the alteration of cholinergic and serotonergic pathway genes affecting cell proliferation. (PubMed, Sci Rep)
Collectively, these data indicate that submicromolar DZN promotes proliferation and suppresses apoptosis of GC cells by perturbing neuronal signaling networks. Our findings provide novel evidence linking environmental OP exposure to dysregulated neurotransmitter pathways in gastric carcinogenesis and highlight neuronal signaling components as potential therapeutic or preventive targets.

The molecular footprint of the serotoninergic system in human glioblastoma cells. (PubMed, J Mol Histol)
Additionally, by using RT-qPCR and immunofluorescence, we observed a differential expression and intracellular distribution pattern of all serotoninergic system elements in human glioblastoma-derived cell lines. Our findings demonstrate the differential expression and cellular localization of the serotoninergic elements in human glioblastomas and suggest their potential participation in their progression.
