Additionally, BBR preserved cellular proliferation, decreased TUNEL+ apoptotic cells and the inflammatory response in SMG tissues and organoids in HNR-induced xerostomia models. In conclusion, this study demonstrates that BBR can increase saliva secretion in healthy and HNR mice, indicating its potentiality for the treatment of radiation-induced xerostomia.

Identification of tumor initiating cells and early marker genes in normal colonic epithelium that lead to neoplastic transformation. (PubMed, Res Sq)
Conclusions : This study identifies TICs as the developmental origin of neoplastic stem-like states and delineates early transcriptional and pathway reprogramming events that drive the transition from normal to premalignant colonic epithelium. These findings provide new insight into CRC initiation and nominate biomarkers with translational potential for early detection and therapeutic targeting.

2 mM NaVPA-3 mM NaDCA, as well as temozolomide, had individual impacts on the SLC12A2, SLC12A5, CDH1, CDH2, EZH2, and GFAP expressions of tested GBM5-1, GBM5-2F, and GBM5-3F primary cells of female GBM patients. The combination of 2 mM NaVPA-3 mM NaDCA may have an advantage in antitumor activity and may be more effective than TMZ; however, the effect is individual.

Gut microbiota dysbiosis (enrichment of nitrogen-cycle bacteria and depletion of short-chain fatty acids producers) induces chromosomal instability through the ammonium ion-solute carrier family 12 member 22 (SLC12A2)-NEK2 axis. Therefore, risk-based screening among high-risk populations (e.g., those who are obese, elderly, or chemically exposed), along with early interventions targeting cytogenetic abnormalities [e.g., B cell lymphoma 2 (Bcl-2) inhibitors for t(11;14), ferroptosis inducers for t(4;14)] and modifiable factors (e.g., vitamin D supplementation, gut microbiota modulation), may effectively delay disease progression and improve prognosis.

Collectively, our results indicate that prolonged afatinib treatment enhances cAMP-induced fluid secretion by mechanisms involving EPAC-TMEM16A-Kv7.1-mediated fluid secretion and secretory cell differentiation in 3D mouse mini-gut models. The EPAC-TMEM16A-Kv7.1-mediated fluid secretion represents a promising therapeutic target for treating afatinib-induced diarrheas.

Evaluation of the Choroid Plexus Epithelium Inflammation TLR4/NF-κB/NKCC1 Signal Pathway Activation in the Development of Hydrocephalus. (PubMed, CNS Neurosci Ther)
These results confirmed that the inflammatory reaction contributes TLR4/NF-κB/NKCC1 mediated CPE abnormal secretion and consequent hydrocephalus. Regulation of TLR4/NF-κB/NKCC1 and AQP1 can prevent this process. Our study provides a strong rationale for further exploring alleviating CPE abnormal secretion as a therapeutic perspective of hydrocephalus.
