Exceptional long-term survival in pulmonary large-cell neuroendocrine carcinoma with brain metastases: a case report. (PubMed, Front Oncol)
This case demonstrates that prolonged survival in stage IV LCNEC with brain metastases is possible through individualized multimodal therapy and close longitudinal monitoring. It underscores the potential value of molecular profiling and biomarkers like PRO-GRP in treatment planning and disease tracking.

A Novel Targeted Sequence for Chromosome 11p15.5 Maternal Loss in SDHD-Related Paragangliomas. (PubMed, Genes Chromosomes Cancer)
In our studies, 12/13 SDHD-related tumors demonstrated complete loss of the maternal 11p15.5-15.4 region, preventing localization of a specific driver TSG. Only one exceptional SDHD-related tumor retained this region, warranting further investigation into the mechanism underlying parent-of-origin tumorigenesis.

An Unusual High-Grade Sarcoma of the Kidney with TPM4::NTRK1 Fusion, CDKN2A/B Deletion, SDHD and NOTCH3 Variants: The First Case Report in Pediatric Population. (PubMed, Fetal Pediatr Pathol)
Based on these molecular findings, the diagnosis was confirmed as an NTRK-rearranged high-grade sarcoma with an unusual histomorphological phenotype. This case highlights novel molecular characteristics of NTRK-rearranged high-grade sarcoma and underscores the critical role of comprehensive molecular profiling in diagnosing challenging cases.

Transcriptional subtypes on immune microenvironment and predicting postoperative recurrence and metastasis in human pheochromocytoma and paraganglioma. (PubMed, Elife)
Notably, we identify ANGPT2, PCSK1N, and GPX3 as key subtype-specific biomarkers, with ANGPT2 driving tumor progression in C1 and emerging as a potential therapeutic target. Our findings provide a refined molecular classification integrating immune and genomic features in human PPGLs, offering a framework for improved prognostication and precision therapies in this rare neuroendocrine tumor type.

Phenotypic Carney-Stratakis syndrome with DIS3L2 variant: a case challenging the current genetic paradigm. (PubMed, Proc (Bayl Univ Med Cent))
The patient has been on imatinib since July 2023 with controlled disease. To the best of our knowledge, this is the first reported case of CSS with a DIS3L2 variant. Further reports and studies are needed to establish a definitive association between this gene and CSS.

French recommendations on multi-gene panel testing in renal cell carcinoma. (PubMed, Eur J Med Genet)
Unlike hereditary predisposition to breast or colon cancer, hereditary renal cancer predispositions are rare syndromes and risk estimates are lacking for most of them. Prospective studies are needed to improve our knowledge. The GGC-PREDIR experts retained 12 genes for inclusion in the NGS MGP for renal cancer patients. However, the panel will be expanded on the basis of regularly updated data from the medical literature.

Metastatic disease in phaeochromocytomas and paragangliomas in various genotypes - A systematic review and meta-analysis. (PubMed, J Clin Endocrinol Metab)
We present associations between genotype and metastatic disease in PPGL patients. Our results indicate that patients harbouring a pathogenic genetic variant have a higher rate of metastases compared to patients with no identified variant. High heterogeneity in several analyses suggests that further large cohort studies are needed.
