Glycolysis reprogramming predicts poor prognosis and drives therapy resistance via CLN6 in lethal prostate cancer. (PubMed, Acta Biochim Biophys Sin (Shanghai))
In vitro, enzalutamide-resistant prostate cancer cell lines exhibit heightened glycolysis, and 2-DG inhibition reverses this effect, restoring drug sensitivity. CLN6 knockdown reduces glycolytic activity and cell proliferation. The GLY score offers robust prognostic value, and CLN6 represents a promising therapeutic target for precision medicine in lethal prostate cancer.

Phenotypic Carney-Stratakis syndrome with DIS3L2 variant: a case challenging the current genetic paradigm. (PubMed, Proc (Bayl Univ Med Cent))
The patient has been on imatinib since July 2023 with controlled disease. To the best of our knowledge, this is the first reported case of CSS with a DIS3L2 variant. Further reports and studies are needed to establish a definitive association between this gene and CSS.

Unlike hereditary predisposition to breast or colon cancer, hereditary renal cancer predispositions are rare syndromes and risk estimates are lacking for most of them. Prospective studies are needed to improve our knowledge. The GGC-PREDIR experts retained 12 genes for inclusion in the NGS MGP for renal cancer patients. However, the panel will be expanded on the basis of regularly updated data from the medical literature.

Exploring the Glycolytic Mechanisms in "Driver Gene-Negative" Lung Adenocarcinoma (LUAD): A Single-Cell RNA Sequencing Approach to Identify the MIF-HIF-1α Axis. (PubMed, J Cancer)
Single-cell sequencing analysis validated the key role of MIF in the glycolysis process and revealed a positive feedback regulatory axis between MIF and HIF-1α, which may promoting glycolysis and malignant transformation. This study elucidated glucose metabolic reprogramming mechanisms and highlighted the MIF-HIF-1α axis as a promising therapeutic target in "driver gene-negative" LUAD, which may offer new avenues for improving outcomes, particularly those lacking conventional targeted therapy options.

Metastatic disease in phaeochromocytomas and paragangliomas in various genotypes - A systematic review and meta-analysis. (PubMed, J Clin Endocrinol Metab)
We present associations between genotype and metastatic disease in PPGL patients. Our results indicate that patients harbouring a pathogenic genetic variant have a higher rate of metastases compared to patients with no identified variant. High heterogeneity in several analyses suggests that further large cohort studies are needed.

Discovery of KMT5A repressed miR-99b cluster with potential to restore chemotherapy sensitivity in gastric cancer by regulating mitochondrial Complex II and affecting OXPHOS. (PubMed, Pharmacol Res)
Findings from GC cells, organoids and PDX models demonstrated that overexpression of the miR-99b cluster sensitized GC to cisplatin, likely through its inhibitory effects on mitochondrial respiratory function, particularly OXPHOS...Moreover, elevated KMT5A expression and decreased miR-125a-5p expression indicated both poorer prognosis and chemo-resistance in patients with GC. This study highlights the multifaceted roles of the miR-99b cluster in GC and offers novel perspectives for the development of innovative therapeutics aimed at overcoming chemoresistance and enhancing treatment efficacy for GC patients.

Mitochondrial electron transport chain gene-based prognostic model identifies SDHB as a key regulator of low-grade glioma progression and therapeutic target. (PubMed, Cancer Cell Int)
This study provides a comprehensive analysis of METCGs in LGG and develops a robust prognostic model for patient stratification. SDHB, a key subunit of Complex II, plays a crucial role in mitochondrial function and tumor progression. Our findings suggest that he high expression of SDHB in LGG contributes to maintaining elevated SDH and Complex II activity, ensuring the structural and functional integrity of mitochondrial ETC complexes. This supports the high ROS production and MMP required for the rapid growth of LGG, thereby promoting its proliferation and invasion. Thus, targeting SDHB and its associated pathways could offer new therapeutic avenues for LGG treatment.

Succinate dehydrogenase-related PPGL: Clinical and Genetic Insights from Indian Study. (PubMed, Endocr Connect)
