Succinate Dehydrogenase-Deficient Cancer Cells Have Increased Susceptibility to Ym155-Induced DNA Damage. (PubMed, Endocr Relat Cancer)
In summary, accumulation of succinate in SDH-deficient tumors inhibited KDM4 activity, impaired DNA repair and yielded enhanced susceptibility to Ym155-induced reactive oxygen species (ROS) generation. The identified intrinsic susceptibilities of SDHB-deficient cancers have the potential to be therapeutically leveraged.

In this issue of the JCI, Lee et al. have described a newly developed cellular complementation assay for SDHB function that may assist variant curation in clinical practice and thereby improve outcomes for patients inheriting these cancer-risk variants.

S100 Expression and SDHB Status Define Distinct Biological Subsets of Bladder Paraganglioma. (PubMed, Hum Pathol)
These findings demonstrate that diffuse S100 positivity in bladder paraganglioma is relatively common and represents a significant diagnostic pitfall. Importantly, S100-positive tumors appear to be associated with intact SDHB expression and a potentially less aggressive phenotype, whereas SDHB loss identifies a higher-risk subset with increased metastatic potential.

Clinicopathological and molecular features of acquired cystic disease-associated renal cell carcinoma (PubMed, Zhonghua Bing Li Xue Za Zhi)
ACD-RCC is a rare renal cell carcinoma that occurs in patients with end-stage renal disease and has unique morphological features. It is often associated with favorable prognosis and alterations in genes related to the MTOR/TSC pathway or chromatin modification.

Bladder Paraganglioma With Rapid Metastasis: A Case Report and Review of the Literature. (PubMed, Cureus)

Overall, metabolic RCC recognition in Italy is primarily morphology-driven but constrained by uneven access to confirmatory IHC, particularly 2SC, and to molecular assays. The findings argue for harmonized diagnostic algorithms, regional reference laboratory networks, and routine involvement of molecular tumor boards, supported by targeted educational initiatives (including curated digital slide repositories), to standardize practice and improve patient pathways from morphologic suspicion to genetic counselling and tailored surveillance.

Elucidation of gene clusters underlying withanolide biosynthesis in ashwagandha through yeast metabolic engineering. (PubMed, Nat Plants)
Identifying SULF1 as a core pathway enzyme challenges the conventional paradigm of sulfotransferases as tailoring enzymes and suggests a wider role for this enzyme family in plant specialized metabolism. This work opens new avenues for the sustainable production of withanolides through biomanufacturing and for drug development leveraging the withanolide scaffold.

The identification of a rare VHL c.369delG variant, previously unreported in association with PPGL, within a characteristic VHL-related clinical phenotype highlights the importance of this association. Succinate dehydrogenase B palmitoylation promotes T cell exhaustion through the H3K27ac-PD1 axis in pancreatic cancer. (PubMed, Cancer Lett)
As a competitive inhibitor, CPP-S1 inhibits the palmitoylation modification of SDHB, reduces the SDHB protein level, and suppresses the tumor growth of pancreatic cancer cells, with enhanced effects when used in combination with anti-PD1 therapy. These findings reveal a new strategy to overcome the immune escape mediated by the DHHC5-SDHB axis in pancreatic cancer immunotherapy.
