The independent prognostic factors identified in this study provide a theoretical basis for individualized prognosis judgment in OCCC and HGSC. The SMGs and TMB levels may serve as valuable indicators for prognosis and evaluating targeted therapy or immunother-apy efficacy. Druggable genes such as NOTCH1 and RYR3 offer promising therapeutic tar-gets, while stage-specific pathway enrichments reveal potential intervention strategies. Further validation in larger cohorts is needed to confirm these findings. Our study advances the under-standing of molecular heterogeneity in ovarian cancer and lays the groundwork for personal-ized treatment strategies, ultimately improving patient outcomes.

Aberrant DNA hypermethylation-regulated long non-coding RNA RYR3-DT affects ferroptosis and progression of glioma cell through the CoQ10/FSP1 axis. (PubMed, Discov Oncol)
Western blot analysis and RNA sequencing proved that RYR3-DT regulated the ferroptosis CoQ10/FSP1 pathway, concluding that lncRNA RYR3-DT deletion might suppress glioma cell growth migrating and invading ability, and also increase cellular apoptosis via CoQ10/FSP1 pathway. Our research recommends lncRNA RYR3-DT is an expected target for future discoveries on glioma treatment.

Artificial intelligence-based epigenomic, transcriptomic and histologic signatures of tobacco use in oral squamous cell carcinoma. (PubMed, NPJ Precis Oncol)
Using The Cancer Genome Atlas (TCGA) cohort (n = 257) and an internal cohort (n = 40), we identify 3 epigenetic markers (GPR15, GNG12, GDNF) and 13 expression markers (IGHA2, SCG5, RPL3L, NTRK1, CD96, BMP6, TFPI2, EFEMP2, RYR3, DMTN, GPD2, BAALC, and FMO3), which are dysregulated in OSCC patients who were never smokers vs. those who have a ≥ 10 pack year history. While mortality risk prediction based on smoking status and clinicopathologic covariates alone is inaccurate (c-statistic = 0.57), the combined epigenetic/expression and histologic signature has a c-statistic = 0.9409 in predicting 5-year mortality in OSCC patients.

Genomic Alterations as Independent Prognostic Factors to Predict the Type of Lung Cancer Recurrence. (PubMed, Gene)
Our findings indicate distinct genomic signatures in the LR and DR cohorts, with microtubule disturbance and calcium regulation playing a crucial role in invasiveness in DR of LC. Understanding molecular mechanisms of LC recurrence may lead to the discovery of novel drug targets that could potentially stop spread of cancer cells.

Muscle and Muscle-like Autoantigen Expression in Myasthenia Gravis Thymus: Possible Molecular Hint for Autosensitization. (PubMed, Biomedicines)
Genes encoding autoantigens that share epitopes with AChR-α (NEFM and HSP60), RYR1 (neuronal RYR3), and TTN (NEFM) were up-regulated in thymomas versus hyperplastic and control thymuses, with distinct molecular patterns across the thymoma histotypes that could be relevant for autoimmunity development. Our findings support the idea that altered muscle autoantigen expression, related with hyperplastic and neoplastic changes, may favor autosensitization in the MG thymus, and that molecular mimicry involving tumor-related muscle-like proteins may be a mechanism that makes thymoma prone to developing MG.
