Clinicopathological and functional evaluation of replication protein A in epithelial ovarian cancers: A target validation study. (PubMed, Transl Oncol)
Platinum resistant ovarian cancer cells were depleted for RPA 1 or 2 and tested for cisplatin, olaparib and talazoparib sensitivity. HAMNO monotherapy was cytotoxic to sensitive and resistant cells. We conclude that RPA directed precision oncology strategy could be a viable strategy in HGSOC.

HAT1 as a Lactyltransferase to Promote DNA Repair Through RPA1 Lactylation in Glioblastoma. (PubMed, Int J Biol Sci)
Correspondingly, targeted inhibition of RPA1 lactylation at K88, using a competitive peptide, reverses radioresistance in GBM cells. Collectively, these findings establish the lactate-driven HAT1-RPA1 lactylation axis as a critical regulator of radioresistance and nominate both HAT1 and lactylated RPA1 as novel therapeutic targets in GBM.

Ascites circRNA ASCOR Drives Platinum Resistance of High-Grade Serous Ovarian Cancer by Facilitating RPA1 Nuclear Translocation. (PubMed, Adv Sci (Weinh))
Targeting ASCOR with antisense oligonucleotides reverses chemoresistance in mouse models. Our study unveils an ascites circRNA-driven pathway underlying platinum resistance in HGSOC, suggesting targeting ASCOR is a promising strategy for HGSOC treatment.

Condensins regulate resection-dependent DNA double-strand break repair pathways in replicated chromatin. (PubMed, Nucleic Acids Res)
Cytogenetic analysis revealed inhibition of chromosome break repair and visible chromatin decondensation, suggesting that condensins function to maintain an appropriate chromatin state for efficient DSB repair in G2-phase. These results identify for the first time condensins as G2 phase-specific regulators of genome stability by fine-tuning HR and other resection-dependent DSB repair pathways.

Orf Infection Localized in the Earlobe: The First Case Report in the Literature (PubMed, Mikrobiyol Bul)
In conclusion, it should be kept in mind that orf lesions may occur even in unexpected sites such as the periauricular region, particularly in patients living in rural areas with a history of animal contact. Early diagnosis of orf infection will prevent unnecessary aggressive surgical interventions and will also contribute to the proper follow-up of patients and the prevention of complications.

Here, we review recent findings on the roles of SSBPs and their coordination during telomere replication and elongation. We also present recent advances on ssDNA-binding factors and their regulators in alternative lengthening of telomeres (ALTs), highlighting how these findings may provide viable therapeutic targets in ALT cancers.

NKX2-1 regulates cell survival, maturation, and DNA-damage responses as a cofactor of RUNX1 in T-cell acute lymphoblastic leukemia. (PubMed, Haematologica)
Furthermore, NKX2-1 positive cells showed less induction of DNA damage and apoptosis upon treatment with etoposide, a DNA damaging chemotherapy agent that is clinically used to treat T-ALL...Notably, NKX2-1 expressing cells showed higher sensitivity towards RUNX1 inhibition, suggesting a cooperative role in regulating T-ALL cell survival. This work reveals a critical role of NKX2-1 in enhancing T-ALL cell survival through DNA damage protection, and identifies RUNX1 as an important cofactor in T-ALL pathogenesis.

Oncometabolite fumarate impairs ATR-CHK1 signaling by succinating RPA1 in Fumarate Hydratase-deficient renal cell carcinoma cells. (PubMed, bioRxiv)
Our findings establish that fumarate-induced succination of RPA1 impairs DNA repair and cell cycle control, promoting genomic instability in FH-dRCC. This work reveals a novel mechanism by which oncometabolites contribute to genomic instability.

HAT1 functions as a lactyltransferase and mediates RPA1 lactylation to promote DNA repair and radioresistance in lung adenocarcinoma. (PubMed, Cell Death Dis)
Interestingly, we also found that K15 auto-lactylation of HAT1 can modulate its lactyltransferase activity. In conclusion, our research reveals that HAT1-regulated RPA1 lactylation plays an important role in homologous recombination and radioresistance, suggesting that HAT1 may become a potential therapeutic target for reversing the radioresistance caused by lactate accumulation in cancer cells.
