Rbpj deletion in hepatic progenitor cells attenuates endothelial responses and fibrosis in DDC-fed mice. (PubMed, Hepatol Commun)
Our findings reveal Rbpj-dependent expression of reactive cholangiocyte markers in HPCs and demonstrate that Rbpj deletion in HPCs attenuates not only endothelial responses but also liver injury, fibrosis, and VCAM1 expression in cholangiocytes, highlighting the crucial role of HPCs in pathogenic progression.

Our findings imply that downregulation of RBPJ in PDAC patients derepresses Notch targets and promotes KRAS-mediated pancreatic acinar cells transformation and desmoplasia development.

Hsa_circ_0001535 Regulates Colorectal Cancer Progression via the miR-433-3p/RBPJ Axis. (PubMed, Biochem Genet)
In addition, in vivo experiment had shown that circ_0001535 knockdown inhibited tumor growth by up-regulating miR-433-3p and inhibiting RBPJ expression. The circ_0001535/miR-433-3p/ RBPJ axis plays a catalytic role in the progression of CRC, which may provide new insights into the molecular mechanism of CRC.

Therapeutic targeting of RBPJ, an upstream regulator of ETV6 gene, abrogates ETV6-NTRK3 fusion gene transformations in glioblastoma. (PubMed, Cancer Lett)
It was found to be binding with the DNA binding domain of RBPJ with antitumorigenic properties and minimal organ toxicity. Hence, a new target RBPJ, regulating the production of ETV6 and ETV6-NTRK3 fusion protein was found along with a potent RBPJ inhibitor Furamidine.

Exosomal hsa_circ_0004658 derived from RBPJ overexpressed-macrophages inhibits hepatocellular carcinoma progression via miR-499b-5p/JAM3. (PubMed, Cell Death Dis)
Mechanistic investigations revealed that hsa_circ_0004658 acts as a ceRNA of miR-499b-5p, resulting in the de-repression of JAM3. These results indicate that exosome circRNAs secreted from RBPJ Mφ inhibits tumor progression through the hsa_circ_0004658/miR-499b-5p/JAM3 pathway and hsa_circ_0004658 may be a diagnostic biomarker and potential target for HCC therapy.
