Pre-analytical confounders (blood collection tubes, hemolysis, storage conditions), platform-dependent analytical variation, lack of external quality assessment programs, and scarcity of demonstrations of clinical utility in prospective biomarker-directed research are critical translational barriers. Such standardized handling of specimens, commutable reference materials, and reporting of the characteristics of analytical performance would be clearly linked to effective clinical use in accordance with the requirements of laboratory medicine.

Shox2 and Rassf1a DNA methylation: diagnostic utility and association with clinical stage, histological progression and gene mutational landscape in lung adenocarcinoma. (PubMed, Front Oncol)
Combining Shox2 and Rassf1a methylation with EGFR mutations demonstrated enhanced discriminative capacity for early-stage lesions. Our study demonstrated that comprehensive analysis of methylation and gene mutations could provide a novel clinical strategy for molecular subtyping and precision medicine in LUAD.

A scientometric study on methylation modification and identification of related genes in oral potentially malignant disorders and oral cancer. (PubMed, J Dent Sci)
The most frequent methylation related microRNA was miR-296, followed by miR-193 and miR-137. This study for the first time elucidated the scientometric characteristics of all the publications on methylation modification in oral carcinogenesis, and would provide new insights for researchers to comprehend the methylation specific gene profile related OPMD/OSCC.

This regulatory interaction was confirmed through DNA methylation sequencing, methylation-specific PCR, and chromatin immunoprecipitation assays. These findings highlight the pivotal role of DNMT1-driven RASSF1A methylation in facilitating breast cancer metastasis to the brain, and suggest that targeting DNMT1 may represent a promising therapeutic approach for managing metastatic breast cancer.

Genomic Analysis of Low-Grade Serous Ovarian Cancer: Clinical and Biological Insights. (PubMed, Cureus)
The cooperative GOG 281/LOGS trial showed that trametinib, an MEK inhibitor (MEKi), was significantly more effective than standard-of-care options (including chemotherapy or hormonal therapy) in increasing progression-free survival (median PFS 13.0 months vs. 7.2 months; hazard ratio 0.48, p < 0.001)...Genomic and multi-omic profiling have revealed actionable vulnerabilities and precision oncology approaches. The advent of biomarker-directed trials, molecular subtyping incorporation, and innovative computational strategies is likely to gradually ameliorate therapy selection and, thereby, finally improve long-term outcomes for patients with this complex disease.

Structural basis of microtubule-mediated signal transduction. (PubMed, Cell)
Building on this sequestration-and-release mechanism, we identify microtubule-binding C1 domains in additional signaling proteins, including other guanine nucleotide exchange factors (GEFs), kinases, a GTPase-activating protein (GAP), and a tumor suppressor, and show that microtubule-mediated regulation via C1 domains is conserved in the Ras association domain-containing protein 1A (RASSF1A). Our findings establish a structural framework for understanding how microtubules can function as spatiotemporal signal sensors, integrating and processing diverse signaling pathways to control important cellular processes.
