An endothelial RNA splicing atlas catalogs effects of IL-1β and identifies an alternative PROCR isoform with genetic links to pleiotropic vascular disease. (PubMed, Am J Hum Genet)
For example, genetic association for a previously undescribed isoform of endothelial protein C receptor (PROCR) colocalized with genetic risk for deep vein thrombosis and coronary artery disease with opposing risk alleles. This study demonstrates the prevalence of inducible promoters upon inflammatory stimuli and shows that genetic risk for vascular disease may be in part governed through AS in ECs.

Auguries of adaptivity: LES γδ TCR ligand recognition revisited. (PubMed, Trends Immunol)
In a significant advance more than 12 years ago, endothelial protein C receptor (EPCR), a cell-surface-expressed major histocompatibility complex (MHC)-like protein that binds phospholipids, was identified as the first ligand for a human γδ TCR to be validated by direct binding experiments: a finding that undoubtedly posed more questions than it answered. In this review we discuss how features of this single clonotypic specificity anticipated insights into adaptive-like human γδ T cell biology that emerged in subsequent investigations, and we highlight recent findings about EPCR that point towards the relevance of such responses in anti-pathogen and potentially anti-tumour immunity.

Activated protein C promotes human lung cancer progression through the release of tumor extracellular vesicles and transfer of microRNA-200a. (PubMed, Cell Death Dis)
Consistent with these findings, our in vivo studies in BALB/c nude mice showed that aPC-EVs from lung cancer cells promote tumor growth, metastasis, and drug resistance through miR-200a transfer. Targeting EV biogenesis, EV's miR-200a, and/or EV uptake mechanisms may offer novel therapeutic strategies in limiting lung tumorigenesis, thereby increasing patients' survival.

Among the top signals are novel isoforms of Endothelial Protein C Receptor (PROCR) and Distal Membrane Arm Assembly Component 2 (DMAC2), whose splicing patterns colocalize with risk for coronary artery disease (CAD). This study demonstrates the prevalence of inducible alternative promoters and supports that ECs express numerous novel transcripts regulated by genetics and inflammation that are consistent with driving individual risk for cardiovascular disease.
