The rare report of unicentric Castleman disease with concurrent myasthenia gravis and paraneoplastic pemphigus: a case report with a focused review of the literature. (PubMed, Front Oncol)
Accurate recognition required integration of histopathology, immunofluorescence, and serological testing, achieved through multidisciplinary collaboration across thoracic surgery, neurology, dermatology, and pathology. Clinicians should remain vigilant for such complex presentations, as early diagnosis and coordinated management are essential to improving outcomes in rare autoimmune-associated CD.

Plakin Expression in Serous Epithelial Ovarian Cancer Has the Potential to Impede Metastatic Spread and Epithelial-Mesenchymal Transition: A Comparative Expression Analysis of Immunohistochemical and In Silico Datasets. (PubMed, Cancers (Basel))
Our data suggest a partial loss of plakin expression as EOC tumours progress. This may impact the connections of plakins with membrane-bound receptors, which impede the downstream signalling required for the initiation of EMT as the tumours progress.

Plakins are involved in the regulation of centrosome position in polarized epithelial cells. (PubMed, Biol Cell)
These results revealed that the down-regulation of plakins and the consequential centrosome mispositioning are key signatures of disorganised cytoskeleton networks, inter-cellular junction weakening, shape deregulation and the loss of polarity in breast cancer cells. These metrics could further be used as a new readouts for early phases of tumoral development.

Identification of differentially methylated regions associated with both liver fibrosis and hepatocellular carcinoma. (PubMed, BMC Gastroenterol)
Changes in the methylation and transcriptional levels of ZBTB38, ZC3H3, FOXK1, and KAZN are important for the development of fibrosis and HCC; and are therefore potential therapeutic targets and diagnostic tools for cirrhosis and HCC.

Paraneoplastic pemphigus uncovers distinct clinical and biological phenotypes of western unicentric Castleman disease. (PubMed, Br J Haematol)
These features were not found in patients with UCD alone or in the PNP group without UCD. These data indicate that UCD-PNP patients belong to a subgroup sharing strong clinical and biological identity that might help to decipher the different dynamics of UCD natural history.

A case of paraneoplastic pemphigus in association with malignant peripheral nerve sheath tumor (EADV-Sp 2023)
For patients who have blisters and are being diagnosed by MPNST, paraneoplastic pemphigus may be considered. We report this rare case for educational purposes.

Quantitative analysis of DNA methylation using sequence-specific, real-time loop-mediated isothermal amplification. (PubMed, Anal Chim Acta)
Moreover, a higher methylated level of kazrin periplakin interacting protein (KAZN) gene was detected in colorectal cancer cells with a stable housekeeping gene β-actin used as an endogenous control. The developed PSS probe for LAMP provide an option to analyze DNA methylation levels in resource-limited environments, and hold great potential for the diagnosis of methylation-related diseases.

CD103 and periplakin are potential biomarkers for response of metastatic melanoma to pembrolizumab. (PubMed, Melanoma Res)
The density and proportion of CD8+ T cells expressing CD103+ positively correlated with PD-L1 expression, though PD-L1 expression was not significantly correlated with outcomes. This screening study found that increased density and proportion of CD8+ T cells expressing CD103 and decreased density of periplakin were associated with positive outcomes in patients with melanoma metastases treated with pembrolizumab and may warrant further study.
