Structural and Dynamic Insights into Podocalyxin-Ezrin Interaction as a Target in Cancer Progression. (PubMed, J Xenobiot)
Through virtual screening, we identified NSC305787 as a selective destabiliser of the R495W mutant complex by disrupting key Trp495-pre-C-terminal loop Ezrin interactions and causing steric hindrance to PIP2 recruitment. Our findings identified mutation-dependent changes in drug binding that can guide the development and repurposing of compounds for targeting PODXL-related cancers and improve patient outcomes in PODXL-altered malignancies.

Biomimetic core-shell breast cancer models using alginate, gelatin, and collagen I: simulating the tumor matrix for drug evaluation. (PubMed, Int J Biol Macromol)
Breast cancer cells proliferated in the core of all prototypes designed, forming spheroids and cell aggregates with a high resistance to doxorubicin. The addition of Collagen I to the developed model enabled the upregulation of malignancy markers (Col1A1, Ki67, FOXC2, SNAI1, NFKB1, WWTR1), invasion markers (WASL, ACTA1, MYO1E, TPM4, PODXL, ITGA2, ITGA5, MENA, EGFR, CDC42), and drug resistance markers (ABCG2, CYP1A1, BAX, HSP90AA1) occurring in vivo. The developed 3D in vitro model can clarify the contribution of the extracellular matrix to the tumor outcome and drug efficacy by replicating some key characteristics of breast tumors, establishing a novel tool for chemotherapeutic agents and drug screening.

Collectively, these data suggest that PODXL may play an important role in aiding immune evasion in HGSC, at least partly by conferring resistance to NK cell infiltration and the related cytotoxicity.

Luteolin prevents axitinib-induced kidney damage in mice. (PubMed, Chem Biol Interact)
Our results elucidate that axitinib induces podocyte filtration barrier core protein loss, foot process effacement, glomerulosclerosis, and proteinuria through upregulation of TRPC6 protein expression. Additionally, naturally derived luteolin serves as a potential intervention strategy, providing a theoretical basis for the clinical prevention and treatment of nephrotoxicity caused by axitinib and other TKIs.

The Aging Epigenome: Integrative Analyses Reveal Functional Overlap with Alzheimer's Disease. (PubMed, medRxiv)
Finally, we prioritized 9 aging-associated CpGs, located in promoter regions of PDE1B, ELOVL2, PODXL2 , and other genomic regions, that showed strong positive blood-to-brain methylation concordance, as well as association with AD or AD neuropathology in independent studies, after adjusting for age and other covariates. Our findings provided insights into the functional overlap between the aging processes and AD, and nominated promising blood-based biomarkers for future AD research.
