Mechanistically, NUSAP1 governs IP3R phosphorylation to regulate Ca2+ homeostasis, ER stress, and mitophagy, thereby modulating apoptotic signaling in EC. Our study unveils the NUSAP1-IP3R axis as a central driver of EC progression, offering novel therapeutic targets for calcium-dependent oncogenic pathways.

Differentially Expressed Genes Associated with the Development of Cervical Cancer. (PubMed, Int J Mol Sci)
The publicly available microarray datasets, including GSE39001, GSE9750, GSE7803, GSE6791, GSE63514, and GSE52903 in combination with bioinformatics database predictions, were used to identify differential expression genes, potential biomarkers, and therapeutic targets for cervical cancer; additionally, we undertook bioinformatic analysis to determine gene ontology and possible miRNA targets related to our DEGs...Interestingly, hub proteins KIF4A, NUSAP1, BUB1B, CEP55, DLGAP5, NCAPG, CDK1, MELK, KIF11, and KIF20A were found to be potentially regulated by several miRNAs, including miR-107, miR-124-3p, miR-147a, miR-16-5p, miR-34a-5p, miR-34c-5p, miR-126-3p, miR-10b-5p, miR-23b-3p, miR-200b-3p, miR-138-5p, miR-203a-3p, miR-214-3p, and let-7b-5p. The relationship between these genes highlights their potential as candidate biomarkers for further research in treatment, diagnosis, and prognosis.

Discovery of Essential Genes as Possible Targets for Prostate Cancer Drug Development. (PubMed, Int J Genomics)
Following that, we considered these possible genes as targets for drugs, performed docking analysis with 255 meta-drug agents, and identified the top 10 candidate drugs (adapalene, ergotamine, imatinib, dutasteride, vistusertib, risperidone, zafirlukast, irinotecan hydrochloride, drospirenone, and telmisartan). This study uniquely combines multidataset transcriptomic integration, HubG prioritization, and MD validation to propose novel biomarker-drug pairings for PCa. The findings offer promising leads for future experimental and clinical validation.

Based on the drug-likeness, ADME, and toxicity predictions, the 9753624331-0-3 presented favorable properties and hence may be considered the potential lead targeting the NUSAP1 protein.

Prognostic value of fatty acid metabolism-related signature and integrated analysis of the immune microenvironment in multiple myeloma. (PubMed, BMC Cancer)
We identified 37 survival-associated FMGs in MM patients, and verified the effects of CCNA2, KIF11, and NUSAP1 on the cell cycle and proliferation of MM cells. Our results also suggest that survival-associated traits based on these genes are potentially robust prognostic biomarkers for MM patients.

Moreover, NUSAP1 is identified as a novel HIF-repressed gene in TNBC cells, and its expression level shows a negative correlation with clinical outcomes in TNBC patients. These findings establish an HIF-NUSAP1 double-negative feedback loop in TNBC and validate TS-MAD as a potential therapeutic strategy for HIF-driven cancer.

The expression levels of AURKA, DTL, and RRM2 are positively correlated with NUSAP1, indicating a common regulatory relationship among these genes. Our present study demonstrated that AURKA, DTL, NUSAP1, and RRM2 are potential diagnostic and prognostic biomarkers for BRCA, and may also be potential treatment targets for clinical implication in the future.

TRIP13 is upregulated in ccRCC and may serve as a novel prognostic biomarker for patient survival and treatment response. Additionally, TRIP13 enhances ccRCC cell proliferation, invasion, and EMT via the PI3K/AKT/mTOR pathway, while its expression is closely linked to immune cell infiltration and immune checkpoint regulation, offering new insights for immunotherapeutic approaches in ccRCC.

YY1 enhances HIF-1α stability in tumor-associated macrophages to suppress anti-tumor immunity of prostate cancer in mice. (PubMed, Nat Commun)
Either myeloid-specific conditional knockout of YY1 or a treatment with tenapanor for decreasing the YY1-NUSAP1-HIF-1α interaction impairs subcutaneous PCa tumor formation in mouse prostate tumor models. Lastly, a first-generation tetrahedral DNA nanostructure based on the proteolysis targeting chimera technique, termed YY1-DcTAC, allows targeting and degrading YY1 in tumor-associated macrophages for inducing antitumor effects and CD8+ T cell tumor infiltration in mouse tumor models. In summary, our findings underscore the pivotal role of YY1 in the hypoxia/HIF-1α pathway in tumor-associated macrophages and support the targeting of YY1 for treating PCa.
