This MR study establishes plasma miRNAs as causal mediators of gastric carcinogenesis, with miR-409-3p emerging as a key prognostic biomarker. The identified miRNA-target networks highlight actionable pathways for therapeutic intervention, bridging genetic epidemiology with functional genomics in GC precision oncology.

Priorities for validation include reporter assays, gene/protein modulation, phenotypic rescue, and in vivo testing in MP-exposed models. Collectively, our results nominate miRNAs as candidate tools to interrogate and potentially mitigate MP-associated carcinogenic mechanisms.

This preliminary study demonstrated that miR-107, miR-134-5p, miR-149-5p, and miR-370-3p were significantly overexpressed in PCa patients compared to the BPH group. ROC analysis highlighted their diagnostic potential in distinguishing BPH from PCa. Thus, significant metabolomic imbalance in the urine was observed in the OC patients and was associated with changes in the levels of microRNAs that regulate the signaling pathways of the metabolites. The 26 compounds with abnormal concentrations and the levels of the microRNAs miR-33b-5p, miR-423-5p, miR-6843-3p, miR-4668-3p, miR-30c-5p, miR-6743-5p, miR-4742-5p, miR-1207-5p, and miR-17-5p in the urine were considered to be suitable as noninvasive diagnostic markers of OC.

This study suggests that the specific expression of miRNA-1296-5p, miR-499a-5p, miR-374a-5p and miR-370-3p in the serum exosomes of LC, which has diagnostic potential. And the combination of miRNA-1296-5p, miR-499a-5p and miR-370-3p with CEA can further enhance this potential.
