Identification of endometrial cancer biomarkers using weighted gene coexpression network analysis and machine learning. (PubMed, BMC Cancer)
This study found that high MAL expression is significantly correlated with poor prognosis in endometrial cancer patients, which is consistent with its functional role in promoting tumor cell proliferation and invasion. However, after adjusting for clinical factors such as stage and grade, the independent prognostic effect of MAL on overall survival was not prominent, suggesting that it may more likely reflect the extent of tumor progression and adverse clinical features, rather than serving as a sole determinant of prognosis. Therefore, MAL can be regarded as a molecular marker closely associated with disease progression and survival outcomes, and it holds potential for providing auxiliary value in risk stratification and personalized management when combined with clinical parameters.

Atypical manifestation of psoriasis in the elderly: possible expression of Mal de Meleda diagnosis review and case report. (PubMed, Front Med (Lausanne))
Systemic retinoids, particularly acitretin, have demonstrated efficacy in reducing epidermal hyperkeratosis and enhancing clinical outcomes...Distinctive diagnostic clues-such as very early disease onset, autosomal recessive mode of inheritance, and the hallmark transgradient pattern of palmoplantar keratoderma-can aid in distinguishing MDM from the more prevalent psoriasis vulgaris. Although biologic therapies targeting TNF-α and IL-17A have shown limited efficacy in MDM, systemic retinoids continue to represent a clinically effective therapeutic option for managing this rare genodermatosis.

MAL2 and rab17 selectively redistribute invadopodia proteins to laterally-induced protrusions in hepatocellular carcinoma cells. (PubMed, Mol Biol Cell)
We also determined that MAL2 and rab17 interaction was GTP dependent, but not dependent on the MAL2 EVH1 recognition motifs, and that protrusions formed by their combined expression shared features of those induced by either alone. Finally, we report that MAL2 or rab17 can redirect trafficking of newly synthesized membrane proteins from the Golgi to the induced protrusions and that the EVH1 recognition motif was required in MAL2 and that rab17-mediated trafficking was GTP-dependent.

Epsilon Toxin from Clostridium perfringens Induces the Generation of Extracellular Vesicles in HeLa Cells Overexpressing Myelin and Lymphocyte Protein. (PubMed, Toxins (Basel))
We observed that ETX induces the formation of EVs. Moreover, the MAL protein and ETX oligomers are found in these EVs, which are a very useful tool to decipher and study the mode of action of ETX and characterize the mechanisms involved in the binding of ETX to its receptor.

Dietary Intervention with Omega-3 Fatty Acids Mitigates Maternal High-Fat Diet-Induced Behavioral and Myelin-Related Alterations in Adult Offspring. (PubMed, Curr Neuropharmacol)
These findings suggest that Ω3D may play a significant role in correcting behavioral and neurobiological changes caused by adverse prenatal conditions.

ATF1 regulates MAL2 expression through inhibition of miR-630 to mediate the EMT process that promotes cervical cancer cell development and metastasis. (PubMed, J Gynecol Oncol)
ATF1, which modulates the miR-630/MAL2 pathway, affects the EMT process and cervical carcinoma cell growth and spread. Therefore, ATF1 may serve as a promising marker and treatment target for cervical malignancies intervention.

MAL, a potential immunotherapy target, is associated with poor prognosis in cancer patients. (PubMed, Cell Mol Biol (Noisy-le-grand))
MAL was also implicated in immunological pathways in BRCA and LUAD, suggesting the important role of MAL in cancer immune regulation. In conclusion, the pan-cancer study indicates that MAL with excellent prognostic value is a potential immunotherapy target in multiple cancers.

MAL expression downregulation through suppressive H3K27me3 marks at the promoter in HPV16-related cervical cancers is prognostically relevant and manifested by the interplay of novel MAL antisense long noncoding RNA AC103563.8, E7 oncoprotein and EZH2. (PubMed, Clin Epigenetics)
AC103563.8-E7-EZH2 axis, therefore, appears to crucially regulate the expression of MAL, through chromatin inactivation in HPV16-CaCx pathogenesis, warranting therapeutic strategy development.

Research advances of MAL family members in tumorigenesis and tumor progression (Review). (PubMed, Mol Med Rep)
In the present review, an overview of the structural and functional properties of the MAL family and the latest research findings regarding the relationship between several MAL members and various cancers is provided. Furthermore, the potential clinical and scientific significance of MAL is discussed and directions for future research are summarized.

A maternal high-fat diet during pregnancy and lactation induced depression-like behavior in offspring and myelin-related changes in the rat prefrontal cortex. (PubMed, Front Mol Neurosci)
Maternal HFD also induced long-lasting adaptations manifested by the reduction of immature and mature oligodendrocytes in the prefrontal cortex in adult offspring. In summary, maternal HFD-induced changes in myelin-related genes are correlated with depressive-like behavior in adolescent offspring, which persists even to adulthood.

A novel long noncoding RNA located on the antisense strand of MAL promotes the invasion and metastasis of oral squamous cell carcinoma. (PubMed, Arch Oral Biol)
lncRNA-AC103563.8 is a functional lncRNA that promotes OSCC development by acting on MAL or interacting with other tumor-related proteins. This study also indicates that this lncRNA may exert regulatory functions in OSCC and is a potential target for OSCC therapy.
