Comprehensive analysis reveals that LTBR is a immune-related biomarker for glioma. (PubMed, Comput Biol Med)
Moreover, by virtue of the candidate LTBR CpG sites, a prognostic risk-score model was finally constructed to guide the chemotherapy, radiotherapy, and immunotherapy for glioma patients. Taken together, LTBR is closely correlated with immune functions in gliomas, and LTBR DNA methylation could serve as a biomarker for diagnosis and prognosis of gliomas.

Systematic analysis of the prognostic value and immunological function of LTBR in human cancer. (PubMed, Aging (Albany NY))
LTBR is overexpressed in several tumors and is associated with a poor prognosis. It is related to immune-related genes and immune cell infiltration.

Monoclonal Antibodies Targeting LIGHT Block Ltbr Signalling and Eliminate Acute Myeloid Leukemia Stem Cells (ASH 2023)
We recently documented that targeting CD70, the ligand of the tumor necrosis factor receptor (TNFR) CD27, by the antibody-dependent cell-mediated cytotoxicity (ADCC)-enhanced monoclonal antibody (mAb) cusatuzumab eliminates LSC...Our findings reveal that LIGHT/LTbR-signalling is crucial for the pathogenesis of AML and especially for the maintenance and expansion of LSCs. We developed and validated novel LIGHT targeting mAbs in vitro and in vivo for further development in clinical trials.

Silencing of TRAF5 enhances necroptosis in hepatocellular carcinoma by inhibiting LTBR-mediated NF-κB signaling. (PubMed, PeerJ)
Moreover, TRAF5 knockdown suppressed xenograft tumor growth, inhibited cell proliferation, and promoted tumor cell apoptosis. TRAF5 deficiency facilitates necroptosis in HCC by suppressing LTBR-mediated NF-κB signaling.

Preclinical Development of Lymphotoxin Beta Receptor-Overexpressing CAR T Cells Demonstrates Potentiation of CAR T Cell Antitumor Activity (ASH 2022)
A) T cells from a treatment-naïve DLBCL patient were engineered with an anti-CD19 CAR (utilizing the 4-1BB costimulatory domain, based on tisagenlecleucel) co-delivered with LTBR or an irrelevant gene, truncated NGFR (tNGFR)...The study was blinded. Log-rank Mantel-Cox p value is shown.

LTβR overexpression promotes plasma cell accumulation. (PubMed, PLoS One)
LTβR OE PCs had increased expression of Nfkb2 and of the NF-kB target genes Bcl2 and Mcl1, factors involved in the formation of long-lived BM PCs. Our findings suggest a pathway by which Ltbr gene amplifications may contribute to MM development through increased NF-kB activity and induction of an anti-apoptotic transcriptional program.
