LAMA1 derived from colorectal cancer promotes M2 polarization in macrophages via activation of the EGFR/AKT/CREB pathway. (PubMed, Cent Eur J Immunol)
LAMA1 generated from CRC stimulated the EGFR/AKT/CREB signaling pathway in macrophages to induce M2 polarization of macrophages and eventually promote CRC cell proliferation, migration, and invasion, as well as to activate the EMT process and block CRC cell apoptosis. As a pro-carcinogenic factor released by CRC cells, LAMA1 affects the activation of the EGFR/AKT/CREB pathway in macrophages, causing M2 polarization and aggravating the malignant evolution of CRC.

Systems Biology-Based Drug Repositioning Identifies Extracellular Matrix Module as a Therapeutic Target in Lung Squamous Cell Carcinoma. (PubMed, J Med Chem)
In vitro and in vivo experiments validated two repositioned drugs, Fingolimod and Piperlongumine, both targeting ECM components, significantly inhibited LSCC cell growth, proliferation and migration at concentrations below 10 μM. These results provide compelling evidence for the power of systems biology to identify subtype-specific therapeutic vulnerabilities. Our findings highlight a promising framework for precision oncology and underscore the potential of ECM-targeted interventions in LSCC.

Identification of established and novel extracellular matrix components in glioblastoma as targets for angiogenesis and prognosis. (PubMed, Neurogenetics)
Some of these molecules: COL5A1/3, COL6A1, COL22/27A1, FBLN1/2, ITGA3/5, ITGB1 and LAMA1/B1 (p < 0.01); NCAN, HAS1, MMP2/9, TIMP1/2 and TGFB1 (p < 0.05) correlated with GBM patient survival. In conclusion, this study identified both established and novel ECM molecules regulating GBM angiogenesis, suggesting NCAN and COL27A1 are new potential prognostic biomarkers for GBM.

Our advanced ARMS organotypic model mimics (1) the interactions cells-ECM, (2) the cell growth maintenance, and (3) the expression of proteins that characterize tumor expansion and aggressiveness. In the future, the perfusion-based model could be used with primary patient-derived cell subtypes to create a personalized ARMS chemotherapy screening system.

The LAMA1 rs62081531, rs539713, rs566655, and rs607230 polymorphisms were demonstrated to be related to susceptibility to ESCC in the Chinese population. LAMA1 SNPs may have a significant impact on the occurrence of esophageal cancer and may serve as potential diagnostic biomarkers.

A novel risk model based on the correlation between the expression of basement membrane genes and immune infiltration to predict the invasiveness of pituitary adenomas. (PubMed, Front Endocrinol (Lausanne))
Using a variety of bioinformatics methods, we developed a novel risk model to predict the probability of PAs invasion based on the correlation between 11 BM genes and immune infiltration. These findings may facilitate closer surveillance and early diagnosis to prevent or treat IPAs in patients and improve the clinical awareness of patients at high risk of IPAs.
