    Case Report: Rare multisystem metastasis in head and neck paraganglioma with SDHB pathogenic variant and KIF1B VUS manifested as FUO. (PubMed, Front Endocrinol (Lausanne))
    Following multidisciplinary consultation, the patient opted for the cyclophosphamide-vincristine-dacarbazine (CVD) chemotherapy regimen and was subsequently transferred to a regional hospital for coordinated follow-up. A multimodal imaging evaluation and whole-genome sequencing were instrumental in assessing paraganglioma patients. This case suggested that atypical imaging features should raise suspicion of malignant diseases and underscored the importance of interdisciplinary collaboration in guiding the diagnosis and treatment of complex and rare clinical cases.

    Integrated single-cell and transcriptomic analysis of bone marrow-derived metastatic neuroblastoma reveals molecular mechanisms of metabolic reprogramming. (PubMed, Sci Rep)
    Among these, MRPL21 plays a pivotal role in regulating NB cell proliferation and mitochondrial function, underscoring its potential as a promising therapeutic target. These findings suggest that integrated strategies targeting both tumor metabolism and the immune microenvironment may provide new avenues for the treatment of high-risk, metastatic NB.

    CircCCNB1 inhibits vasculogenic mimicry by sequestering NF90 to promote miR-15b-5p and miR-7-1-3p processing in nasopharyngeal carcinoma. (PubMed, Mol Oncol)
    We found that circCCNB1 promoted the processing and generation of miR-15b-5p and miR-7-1-3p through competitive binding to NF90, thereby inhibiting the expression of calumenin (CALU), kinesin family member 1B (KIF1B), RNA polymerase III subunit G (POLR3G), ultimately decreasing the VM of NPC cells. This study not only reveals a new function of circCCNB1 in NPC, but also provides new insights for targeting angiogenesis therapy.

    CX‑5461 potentiates imatinib‑induced apoptosis in K562 cells by stimulating KIF1B expression. (PubMed, Exp Ther Med)
    Moreover, using other leukemic cell lines, it was demonstrated that regulation of KIF1B expression by imatinib/CX-5461 was not a ubiquitous phenomenon in leukemic cells and should be studied in a cell type-specific manner. In conclusion, the results suggested that the synergistic interaction between CX-5461 and imatinib may be of potential clinical value for the treatment of tyrosine kinase inhibitor-resistant chronic myeloid leukemia.

    Screening of Candidate Inflammatory Markers of Epithelial Cells in Hepatocellular Carcinoma Based on Integration Analysis of TCGA/ICGC Databases and Single-cell Sequencing. (PubMed, Recent Pat Anticancer Drug Discov)
    Inflammatory factors CCL7, GPR132, ITGB8, PTAFR, SELL, and VIP influence the inflammatory cascade response in HCC epithelial cells, and their expression correlates with the survival prognosis of HCC patients. Interfering with VIP expression effectively suppresses proliferation, migration, and invasion of HCC cells, as well as inhibiting the occurrence of inflammatory cascade reactions, thus slowing down the progression of hepatocellular carcinoma. Furthermore, compounds NADH and Deferoxamine have the potential to target and bind to the inflammatory protein VIP, highlighting their relevance in potential HCC treatment.

    Malignant Proliferating Pilar Tumor with Sarcomatous Transformation: Case Report with Molecular Profiling (ASDP 2023)
    Shared mutations in carcinomatous and sarcomatous areas include TP53 (previously reported in MPPT), FLT1, HMBS, KIF1B, KMT2D, PML , and RIF1 mutations. CDKN2A nonsense mutation and a higher allelic fraction of a TP53 frameshift mutation are noted only in the sarcomatous component.

    Germline genetic variants in pheochromocytoma/paraganglioma: single-center experience. (PubMed, Endocr Oncol)
    In summary, the landscape of PPGL germline gene variants from 115 Brazilian patients resulted in slightly higher prevalent pathogenic and likely pathogenic variants, especially in the RET gene. We suggest a CJS to identify PPGL patients who would not require initial genetic evaluation, improving test specificity and reducing costs.
