Differential squamous cell fates elicited by NRF2 gain of function versus KEAP1 loss of function. (PubMed, Cell Rep)
These findings demonstrate that, while cellular NRF2 levels are similarly induced, NRF2 gain of function and KEAP1 loss of function elicits distinct fates of squamous cells. The NRF2L30F mutant mouse model developed here will be instrumental in elucidating the mechanistic basis leading to NRF2-activated ESCC.

KEAP1-Mutant Lung Cancers Weaken Anti-Tumor Immunity and Promote an M2-like Macrophage Phenotype. (PubMed, Int J Mol Sci)
We show here that KEAP1-mutant cancers promote immunosuppression of the tumor microenvironment. Our data suggest KEAP1 deletion is sufficient to alter the secretion of cytokines, increase expression of immune checkpoint markers on cancer cells, and alter recruitment and differential polarization of immunosuppressive macrophages that ultimately lead to T-cell suppression.

Impact of STK11 and/or KEAP1 Deletion on Outcomes to Immunotherapy in Non-Small Cell Lung Cancer (IASLC-TTLC 2023)
CONCLUSION STK11 and KEAP1 deletion may be linked to generally worse outcomes to ICI in KRASMUT NSCLC. Future analyses will explore this question in an independent cohort and assess the impact of these deletions on protein expression by immunohistochemistry.

Selective Elimination of NRF2-Activated Cells by Competition with Neighboring Cells in the Esophageal Epithelium. (PubMed, Cell Mol Gastroenterol Hepatol)
Constitutive NRF2 activation promotes the selective elimination of epithelial cells via cell competition, but this competition induces DNA damage in neighboring KEAP1-normal cells, which predisposes them to chemical-induced ESCC.

Nrf2 Activation Sensitizes K-Ras Mutant Pancreatic Cancer Cells to Glutaminase Inhibition. (PubMed, Int J Mol Sci)
This phenomenon was not observed in BxPC3 cells harboring wildtype K-ras. These results indicate the possibility of employing Nrf2 activation and glutaminase inhibition as novel therapeutic interventions for K-ras mutant pancreatic cancers.
