These findings demonstrate that pH-sensitive AKNPs integrate stromal remodeling and metabolic inhibition to synergistically suppress PDAC progression. This nanoplatform provides a promising therapeutic strategy for overcoming stromal and metabolic barriers in pancreatic cancer and may serve as a foundation for future combination regimens.

Immune and Endothelial-Related Extracellular Vesicles Are Associated with Corticosteroid Response and Mortality in Alcohol-Associated Hepatitis. (PubMed, Int J Mol Sci)
EVs enriched in platelet (CD49e) and endothelial (CD31) markers were associated with corticosteroid response, whereas EVs enriched with endothelial (CD105 and CD146) and B lymphocyte (CD19) markers were associated with mortality. Overall, EVs enriched in endothelial and monocyte markers may represent a candidate non-invasive tool for predicting corticosteroid response and mortality in AH, aiding risk stratification and early identification of non-responders for timely transplant evaluation.

Engineered cyclic peptide targeting ITGA5 disrupts tumor-stroma interaction to overcome desmoplasia and resistance in pancreatic ductal adenocarcinoma. (PubMed, Acta Pharm Sin B)
In 3D co-cultured human spheroid models, the peptide decreased markers of resistance (ABCG1, BCL2, CXCR4), stemness (WNT1, CD44) and ECM remodeling (COL1A1, MMP2/9, LOX) and enhanced gemcitabine efficacy...These effects were attributed to reduced desmoplasia, vasculature compression and enhanced infiltration of cytotoxic T cells and apoptosis. This study presents a novel cyclic peptide inhibiting ITGA5-mediated tumor-stroma interaction and thereby reduce desmoplasia and resistance, ultimately enhancing chemotherapy efficacy in PDAC.

Macrophage-associated immune-stromal crosstalks correlate with the tumor microenvironment in microsatellite-stable mucinous colorectal cancer. (PubMed, Pathol Res Pract)
Leveraging cell-type-specific genes and key ligand-receptor pairs, we developed a Mucinous Colorectal cancer Immune Module (MCIM) comprising 18 genes, which stratified patient prognosis and was associated with overall survival in colorectal cancer cohorts. Together, these findings provide a detailed map of the immune-stromal architecture in MAC with MSS status, reveal macrophage-associated immunosuppressive features, and propose MCIM as a potential biomarker for prognostication in mucinous colorectal cancer.

Biomimetic core-shell breast cancer models using alginate, gelatin, and collagen I: simulating the tumor matrix for drug evaluation. (PubMed, Int J Biol Macromol)
Breast cancer cells proliferated in the core of all prototypes designed, forming spheroids and cell aggregates with a high resistance to doxorubicin. The addition of Collagen I to the developed model enabled the upregulation of malignancy markers (Col1A1, Ki67, FOXC2, SNAI1, NFKB1, WWTR1), invasion markers (WASL, ACTA1, MYO1E, TPM4, PODXL, ITGA2, ITGA5, MENA, EGFR, CDC42), and drug resistance markers (ABCG2, CYP1A1, BAX, HSP90AA1) occurring in vivo. The developed 3D in vitro model can clarify the contribution of the extracellular matrix to the tumor outcome and drug efficacy by replicating some key characteristics of breast tumors, establishing a novel tool for chemotherapeutic agents and drug screening.
