Here, a novel combination therapy consisting of the calcineurin inhibitor cyclosporine A (CsA) and the flavone-naphthalimide-polyamine derivative 6c was identified...Finally, combination of CsA and 6c suppressed tumor growth and pulmonary metastasis in vivo. Overall, our study suggests a novel synergistic treatment combination with a comprehensive mechanistic exploration, demonstrating that it is a promising strategy for HCC treatment via targeting RIG-I-like receptor signaling.

The results of analysis viral genes mRNA expression suggested potential role of viral genes during BKV pathogenesis. The results of this limited study showed that at the mRNA level, certain genes of the immune system can be altered and that BKV reactivation has the potential to affect these genes.

Preliminary study on the role of TM9SF2 knockdown in promoting the activity of the type I interferon signaling pathway to inhibit vesicular stomatitis virus replication (PubMed, Xi Bao Yu Fen Zi Mian Yi Xue Za Zhi)
After poly(I:C) stimulation, TM9SF2 knockdown significantly upregulated the mRNA level of IFN-β and the phosphorylation level of IRF3 protein. Conclusion The knockdown of TM9SF2 inhibits the replication of vesicular stomatitis virus, and positively regulates the type I interferon signaling pathway, thus enhancing the host's antiviral innate immune response.

Reconstitution of interferon regulatory factor 7 expression restores interferon beta induction in Huh7 cells. (PubMed, J Virol)
Our study not only identifies a key missing link in the Huh7/Huh7.5 innate immune response but also offers a way to enhance the physiological relevance of these cells in future studies. Our findings pave the way for more accurate modeling of the human hepatic response to viral infections, potentially improving the understanding and management of hepatitis.

Black carp OTUD1 deubiquitinates IRF3/7 and modulates its degradation to negatively regulate the antiviral innate immunity. (PubMed, Fish Shellfish Immunol)
In addition, bcOTUD1 could degrade bcIRF3/7 protein levels, while proteasome inhibitor MG132, lysosome inhibitor chloroquine and autophagy inhibitor 3-MA are able to restore this degradation. Co-immunoprecipitation assay has revealed that bcOTUD1 interacts with bcIRF3/7, and removes K63-linked ubiquitination of bcIRF3 and K48- and K63-linked ubiquitination of bcIRF7. To conclude, our research has brought to light for the first time in teleost that OTUD1 degrade IRF3/7 through decreasing their ubiquitination, thereby restraining IRF3/7-mediated antiviral immune response, which provides an important reference for the study of teleost interferon signaling.

The efficacy of FD9R-GW/siIRF3 in impeding tumor immune evasion was substantiated by an augmented recruitment of cytotoxic T cells and a diminished M2 macrophage polarization in the folate receptor-expressing 4 T1 allograft breast cancer model. Furthermore, the combination of a-PD-1 immunotherapy with FD9R-GW/siIRF3 led to a significant enhancement in the antitumor immune response, as evidenced by the inhibition of circulating tumor-derived exosomal PD-L1.
