Maternal curcumin supplementation alleviates intestinal inflammation of Escherichia coli-infected offspring via modulating gut microbiome in chickens. (PubMed, Anim Nutr)
Concurrently, maternal CUR increased the relative abundance of Lactobacillus (P < 0.05), which was negatively correlated with intestinal inflammation and oxidative stress. The present study showed that maternal CUR supplementation can alleviate intestinal inflammation in offspring chickens, which may be mediated by regulating gut microbiota composition.

Akkermansia muciniphila reduces neuroinflammation and Aβ deposition via tryptophan metabolism in the APP/PS1 mouse model of Alzheimer's disease. (PubMed, Alzheimers Res Ther)
At the same time, A. muciniphila administration improves cognitive deficits, alleviates neuroinflammation and Aβ deposition via AhR/NF-κB/NLRP3 signaling pathway in APP/PS1 mice. In summary, our findings suggest A. muciniphila is a promising approach for preventing AD progression by microbiota-gut-brain axis.

FKBPL-based peptide mimetic, AD-01 (1 nM), in high-glucose conditions, upregulated endothelial vcam1 and glut1 mRNA expression, independent of miR-302b-5p. FKBPL plays an important role in glucose metabolism, endothelial function, angiogenesis, cardiac inflammation and function, and could be explored as a therapeutic target of cardiovascular disease both in nondiabetes and diabetes settings using precision medicine approach.

Phase 1 study of balinatunfib, an oral inhibitor of TNFR1 signal in mild-to-moderate psoriasis. (PubMed, J Eur Acad Dermatol Venereol)
Patients with mild-to-moderate psoriasis treated with balinatunfib reported no severe or serious TEAEs and showed promising clinical responses, suggesting that further evaluation of TNFR1 signal inhibition in inflammatory diseases is warranted.

Relapse of minimal change nephrotic syndrome after initiation of sulfamethoxazole-trimethoprim combination therapy: a case report. (PubMed, CEN Case Rep)
We report an extremely rare case of MCNS relapse triggered by ST combination therapy.A 55-year-old woman with a history of breast cancer treated with tamoxifen developed nephrotic syndrome and was diagnosed with MCNS by renal biopsy...Discontinuation of ST, atorvastatin, and esomeprazole while continuing prednisolone 40 mg/day led to a second remission. Drug-induced lymphocyte stimulation tests for all agents were negative, possibly due to concurrent corticosteroid therapy.Metabolites of sulfamethoxazole have been shown to activate CD4+ T cells and induce multiple cytokines including interleukin-13, interferon-γ, interleukin-22, and granzyme B. Such immune activation could explain the simultaneous occurrence of cutaneous manifestations (drug eruption) and renal relapse (MCNS).Relapse of drug-induced MCNS may occur through either direct podocyte injury or immune-mediated allergic mechanisms. Given the concurrent drug eruption, the latter mechanism appears most consistent with this case.When introducing new medications under immunosuppressive conditions, clinicians should consider the possibility of drug-induced relapse if proteinuria reappears.

Thiophene-based styrene derivative improves colitis symptoms in DSS-induced BALB/C mice through AhR-mediated gut barrier function and inflammatory responses. (PubMed, Toxicol Appl Pharmacol)
TBSD decreased FITC-dextran hyperpermeability, upregulated the tight junction (TJ)-related protein expression levels and regulated the inflammatory mediators including tumor necrosis factor-alpha (TNF-α), interleukin (IL)-6, IL-22 and cyclooxygenase 2 (COX-2) in the Caco-2/RAW264.7 co-culture system and in DSS-induced UC-like mice. Overall, TBSD may be considered as a promising therapeutic agent to improve UC severity through mitigating inflammation, maintaining intestinal mucosal homeostasis and enhancing the intestinal barrier integrity.

Synergistic effects of oral inoculation with recombinant Lactobacillus plantarum NC8 Co-expressing interleukin-2, interleukin-17b and interleukin-26 on infectious bronchitis vaccination in chickens. (PubMed, Poult Sci)
Additionally, viral loads in the vaccinated groups were markedly lower compared to those in the control groups, underscoring the better protective efficacy against the potent IBV strain. In essence, our research indicates that oral administration of NC8-ChIL2/17B/26 augments the overall immune response to the IBV vaccine in chickens, positioning it as a secure and efficacious adjuvant for chicken IBV vaccines.

Cytokine networks in Paneth cell regulation. (PubMed, Mucosal Immunol)
Cytokines such as Interferons (IFNs), Interleukin (IL)-9, IL-13, IL-17, IL-22, and tumor necrosis factor (TNF) influence diverse aspects of Paneth cell biology and, in turn, intestinal tissue homeostasis. In this review, we synthesize current knowledge of cytokine-mediated regulation of Paneth cells, explore the emerging role of Paneth cell phenotyping as predictive in intestinal disease outcome, and conclude with key unanswered questions that define future research directions in the field.

However, the simultaneous decrease in anti-inflammatory cytokines suggests a complex and potentially age-specific immune adaptation to moderate training. Further investigation is warranted to clarify the implications of these immune responses and to refine exercise recommendations for optimal metabolic and immune health in these middle-aged females.

Furthermore, Interleukin (IL)-17f, IL-22, IL-33, and granulocyte colony-stimulating factor (at one month) and C-reactive protein (at three months post-surgery) were positively correlated with delta pain intensity. Our findings support novel cytokine candidates for future research.

Immunomodulatory, antibacterial and antineoplastic potential of Trichilia catigua A. Juss. (Meliaceae) bark extracts and isolated compound cinchonain Ib. (PubMed, Braz J Biol)
Cinchonain Ib presented cytotoxicity to K562 (IC50=46.21±3.9), HL-60 (IC50=51.83±3.17) and MOLT-4 (IC50=57.52±4.57) cells. These results evidenced the availability of uses of T. catigua and cinchonain Ib as immunomodulatory, antineoplastic, bacteriostatic and bactericidal agent.
