M2 macrophage infiltration drives tumor progression and identifies a multigene prognostic signature in esophageal cancer. (PubMed, Front Immunol)
A four-gene M2 macrophage-related prognostic model provides reliable prediction of clinical outcomes in EC. Among these genes, IL1RN and IL36G function as key regulators whose silencing inhibits M2 polarization and attenuates tumor proliferation, invasion, migration, and epithelial-mesenchymal transition.

Association of polycystic ovarian syndrome with inflammatory single nucleotide polymorphism for IL-1 & IL-6 genes: A case-control study. (PubMed, Indian J Med Res)
The heterozygous genotypes of IL-1RN and IL-6 rs1800796 were seen more frequently in the control group (P=0.02), suggesting a potential inverse association. Interpretation & conclusions SNPs in IL-1 and IL-6 genes may influence susceptibility to PCOS and could confer a protective role in women of South Asian Indian origin.

Integrative multi-omics analysis reveals the potential mechanisms of innate immunity in ovarian cancer tumorigenesis and immunotherapy responses. (PubMed, J Ovarian Res)
Molecular docking and molecular dynamics simulations suggest that the Chinese herbal monomer Mulberrofuran K may be a potential therapeutic agent targeting ISG20 for treating OC. In conclusion, we emphasize the significance of innate immunity in OC development and immunotherapy, and find ISG20 to be a prospective biomarker and therapeutic target.

Key genes and associated mechanisms of PCOS and EC comorbidity: A bioinformatics analysis. (PubMed, Medicine (Baltimore))
Collectively, these key genes play a significant role in the occurrence and progression of comorbidities through the pathways mentioned above. The dysregulation of key genes (SYTL1, PARVG, ID4, IL1RN, S100A9) in the context of PCOS-EC comorbidities, along with their associated enrichment pathways, including the TGF-β signaling-pathway and immune microenvironment, plays a significant role in the occurrence and progression of EC.

Transcriptome biomarkers of colon cancer liver metastasis response to neoadjuvant triplet chemotherapy: a case series. (PubMed, J Circ Biomark)
The triplet FOLFOXIRI (fluorouracil, leucovorin, oxaliplatin, and irinotecan) may be considered an effective option in the neoadjuvant setting for metastatic colon cancer (mCRC). Data obtained indicate transcriptional discordance between the primary tumors and liver metastases during neoadjuvant FOLFOXIRI, with the pattern of DEGs involved in their response being distinct. The EPS8L2 transcript could be regarded as a candidate biomarker of liver complete response; however, prognostic conclusions cannot be drawn from this cohort.

Developing models for the diagnosing of ulcerative colitis and prognosis of anti-TNF-α non-response based on neutrophil extracellular trap-associated genes. (PubMed, Front Immunol)
This study aims to identify key NET-associated molecular signatures in UC, develop diagnostic models based on NET-related biomarkers, and construct predictive models for response to anti-TNF-α therapies (infliximab and golimumab). A distinct NET-associated cluster was identified among patients with UC, exhibiting non-responsiveness to anti-TNF-α treatment. Diagnostic and prognostic models based on NET-associated genes hold promise for guiding clinical treatment strategies.

Decoding NMDAR encephalitis: proteomic markers and computational identification of potential therapeutic pathways. (PubMed, Brain Behav Immun)
This study integrates genetic epidemiology with functional validation, successfully identifying protein biomarkers associated with NMDAR-E risk. The convergence of MR-predicted causal proteins with antibody-driven expression changes in human microglia and mouse models underscores their pathological relevance, providing actionable insights for biomarker screening and therapeutic development.

Correlations Between Immuno-Inflammatory Biomarkers and Hematologic Indices Stratified by Immunologic SNP Genotypes. (PubMed, J Clin Med)
IL1RN rs1149222 and TNF-related rs2071645 generate two independent inflammatory signatures-an IL-1β-oxidative axis linked to mild erythropoietic suppression and a TNF-lipid axis without hematologic shift. Integrating targeted genotyping with inexpensive hematologic ratios may refine early risk stratification and guide tailored preventive strategies in ostensibly healthy populations.

The SARS-CoV-2 trigger highlights host interleukin 1 genetics in Epstein-Barr virus reactivation. (PubMed, Cell Rep)
High levels of anti-VCA IgA serve as a strong marker of recent EBV reactivation, which is associated with objective long-term pulmonary dysfunction in LC. Our data highlight the association between host IL1 genetics and EBV reactivation.
