IL-6 as a driver of bone invasion in IFIT2-depleted oral squamous cell carcinoma. (PubMed, Cancer Immunol Immunother)
Notably, a high IL-6/IFIT2 ratio was associated with reduced median survival among OSCC patients. These findings indicate that IFIT2 depletion enhances OSCC-induced bone invasion predominantly through IL-6-mediated osteoclast activation, establishing the IL-6/IFIT2 axis as a critical regulator of OSCC progression and metastasis.

SPIB suppresses protective autophagy via the IFIT2/PINK1/Parkin axis to promote anoikis in colorectal cancer. (PubMed, Cancer Cell Int)
Our results demonstrate that SPIB exerts tumor-suppressive effects during CRC invasion and metastasis through the IFIT2/PINK1/Parkin axis. This study highlights SPIB as a potential therapeutic target for overcoming anoikis resistance in CRC therapy.

Stimulator of interferon genes signaling network-driven prognostic signature for pancreatic cancer: Hub gene discovery with multimodal validation. (PubMed, Oncol Lett)
Subsequent functional experiments involving knockout of IFIT2 and overexpression of ZDHHC1in PAAD cell lines demonstrated that these genes significantly regulated cell proliferation (assessed by CCK-8 assay) and apoptosis (assessed by TUNEL assay) (P<0.001). In summary, the STING signaling pathway is critically involved in PAAD pathogenesis, and the representative genes IFIT2 and ZDHHC1 exhibit distinct functional roles within this context.

BLID is a drug-responsive target of FOXO3a and multi-omics analysis reveals survival mechanisms and therapeutic vulnerabilities in BLID-deficient breast cancer cells. (PubMed, Oncol Lett)
The results of the present study demonstrated that BLID is a target of FOXO3a, and BLID-deficiency in breast cancer cells was associated with modulation of cell death, survival and proliferation, chemoresistance, drug potency, and the interferon response. These findings highlighted BLID as a promising biomarker of drug response and offer a novel framework of integrative mechanisms of therapeutic resistance and disease progression.

Genome-wide analysis reveals the MORC3-mediated repression of PD-L1 expression in head and neck cancer. (PubMed, Front Cell Dev Biol)
MORC3 knockdown also increased the expression of cellular proliferation-related genes and promoted cancer cell proliferation. The present study demonstrated that MORC3 regulates IFN-associated pathways and is a novel repressor of PD-L1 expression and cancer cell proliferation.

Moreover, the expression of IFIT1 and IFIT2 was both negatively related to the levels of TNF-α (r = -0.5027, p < 0.0001; r = -0.4218, p < 0.0001), IL-1β (r = -0.3349, p = 0.0002; r = -0.4070, p < 0.0001) and IL-6 (r = -0.2734, p = 0.0030; r = -0.3536, p < 0.0001), respectively. IFIT1 and IFIT2 can serve as the diagnostic markers for sepsis-related liver injury, and IFIT1 and IFIT2 may participate in the pathological process of sepsis-related liver injury by regulating inflammation and liver function.
