Functional validation demonstrated GRN's critical role in driving epithelial-mesenchymal transition in vitro and conferring resistance to enzalutamide (ENZ) in patient-derived organoids...Furthermore, we identified three novel stromal populations [decorin (DCN)+ endothelial cells, C-C motif chemokine ligand 7 (CCL7)+ fibroblasts, and interferon-induced protein with tetratricopeptide repeats 1 (IFIT1)+ neutrophils associated with disease relapse. These findings illuminate the tumor-immune crosstalk underlying treatment resistance and unveil promising therapeutic targets for overcoming lineage plasticity-driven resistance in advanced prostate cancer.

SYNOPSIS: Clostridium sporogenes alleviates acute and chronic liver injury and inflammation through its metabolite 5-aminovaleric acid (5-AVA). 5-AVA suppresses M1 macrophage activation by upregulating IFIT1, inhibiting the NF-κB pathway, and exerting hepatoprotective effects.

Promotion of hepatocellular carcinoma by small nuclear ribonucleoprotein polypeptide F through upregulation of transmembrane P24 trafficking protein 2 and triggering of the cGAS-STING pathway. (PubMed, Int J Biol Macromol)
In vivo experiments using an orthotopic xenograft tumor model in mice further confirmed that the SNRPF/TMED2/cGAS-STING axis significantly promotes the growth and progression of HCC tumors. In conclusion, this study elucidates the pivotal oncogenic function of the SNRPF/TMED2/cGAS-STING signaling axis in HCC, providing novel strategies for prognosis prediction and targeted therapy.

Mechanistically, NK cell-mediated innate allorecognition drives this early intra-allograft specific neutrophil phenotypic programing.
