Furthermore, by targeting genes that control calcium signaling, cAMP, and inflammation-related pathways, including BCHE, HTR2B, PLA2G2C, PTGER3, and TACR1, this intervention may have the potential to suppress tumor malignancy. By combining multilevel data, this study uncovers the fundamental mechanism of the metabolism-immunity interaction in cervical cancer and suggests a novel approach for hormone-targeted therapy. Our preliminary results indicate that this treatment can upregulate or downregulate certain genes associated with malignant tumors at the mRNA level.

The aggressive colorectal cancer subtype marker HTR2B has a dual role depending on the tumor microenvironment. (PubMed, Cell Commun Signal)
Unfavorable conditions, applying the clinically used 5-fluorouracil, co-culturing with fibroblasts, and collagen-I accumulation in the extracellular matrix (ECM) increased the number of HTR2B + cells...Furthermore, blocking NOTCH activity inhibited the serotonin-induced invasion. Collectively, our results show that the serotonin receptor HTR2B marks a CRC cell population with increased proliferative capacity, invasive potential, and partial EMT, and they highlight the critical role of ECM in shaping the effect of serotonin in CRC.

Gastric Inflammation Impacts Serotonin Secretion in a Mouse Model of Helicobacter pylori Vaccination. (PubMed, Int J Mol Sci)
Analysis of the gastric microbiota revealed that while vaccination did not significantly affect the diversity of the microbiota, vaccinated animals had increased abundance of Lactobacilli. Our results suggest that local inflammation caused by H. pylori is responsible for increased 5-HT release.

Single-cell sequencing of CD45+ cells reveals that HTR2B activation enhances the production of OASL1+ macrophages, contributing to the observed enhancement of antitumor immunity. These findings propose HTR2B as a novel therapeutic target for treating osteosarcoma, offering a dual mechanism of action that directly impedes tumor cell proliferation and augments the host immune response.

Serotonin Influences Insulin Secretion in Rat Insulinoma INS-1E Cells. (PubMed, Int J Mol Sci)
Also, the expression level of the Bcl-2 protein was significantly increased by 50 and 100 nM serotonin treatment compared to the untreated control cells, whereas the levels of Cu/Zn-SOD and Mn-SOD proteins decreased. These results indicate that nanomolar serotonin treatment regulates the insulin protein level but decreases this level in media through membrane vesicle trafficking-related proteins (Rab5, Rab3A, syntaxin6, clathrin, and EEA1), the Akt/ERK pathway, and Htr2b/Htr3a in INS-1E cells.

Targeting MDSC-HTR2B to Improve Immune Checkpoint Inhibitors in Breast to Brain Metastasis. (PubMed, bioRxiv)
In vivo murine models of metastatic and intracranial breast tumors treated with FDA-approved, anti-psychotic HTR2B antagonist, clozapine, combined with immunotherapy anti-PD-1 demonstrated a significant increase in survival and increased T cell infiltration. Collectively, these findings reveal a previously unknown role of MDSC-HTR2B in breast-to-brain metastasis, suggesting a novel and immediate therapeutic approach using neurological drugs to treat patients with metastatic breast cancer.

HTR2B as a novel biomarker of chronic obstructive pulmonary disease with lung squamous cell carcinoma. (PubMed, Sci Rep)
Compared with that in bronchial epithelial cells, HTR2B expression was upregulated in LUSC cells, and inhibiting HTR2B expression led to the inhibition of LUSC cell proliferation. In conclusions, HTR2B might be a new biomarker and therapeutic target in COPD patients with LUSC.

More routinely available TME, SIR and tumor markers alone and in combination deliver reliable prognostic data for choosing the patients with higher risk for propagation. CMS4 is linked with high TSR and poor prognosis, but in overall, CMS-classification showed only limited effect on SIR- and tumor-markers.
