The Role of GRP78/ATF6/IRE1 and Caspase-12 Signaling Pathways in the Protective Effects of n-Hexane Oil Extract of Black Soldier Flies' Larvae (Hermetia illucens) Against Aflatoxin-Induced Hepatotoxicity in Rats. (PubMed, Iran J Pharm Res)
Our histological results showed that BSFL extract (360 mg/kg) could reduce steatosis, cellular swelling, and lobular inflammation induced after AF exposure. The results of this study indicate that BSFL extract, as a valuable bioactive substance with antioxidant properties, may attenuate biochemical indices, oxidative stress, and inflammation caused by AF-induced hepatotoxicity.

β-Hydroxy-β-methylbutyrate attenuates sepsis-associated lung injury by regulating NF-κB p65-mediated inflammation, ER stress and mitochondrial apoptosis in a rat model. (PubMed, Naunyn Schmiedebergs Arch Pharmacol)
HMB protects lungs in experimental sepsis by inhibiting NF-κB inflammation, reducing ER and mitochondrial apoptosis, and boosting antioxidant defenses via NRF2/GPX4. These findings support its potential as adjunct therapy for sepsis-induced ALI.

Two of these DEPsHSPA5 and CAV1were also among the top 50 proteins showing the highest degrees of protein-protein interaction. The DEPs enabled classification of the LUAD patients into three subtypes; the clinical validity of the current subtypes will need to be substantiated in the future with additional LUAD samples and experimentation.

Investigating the correlation of glucose-regulated protein 78 with sperm motility and kinematic parameters insights into male fertility. (PubMed, J Anim Sci Technol)
However, GRP78 expression did not exhibit a significant correlation with progressive sperm motility, average path velocity, curvilinear velocity, straight-line velocity, and amplitude of lateral head displacement. These results provide novel insights into the correlation between GRP78 and sperm motility, as well as its potential implications for male fertility.

RNF26 regulating tumor immunogenicity of hepatocellular carcinoma by degrading GRP78 and instigating ER stress. (PubMed, Drug Resist Updat)
RNF26 is a pivotal molecular node linking the inflammatory microenvironment to immune suppression in HCC. By ubiquitinating and degrading GRP78 to instigate ER stress, it dually regulates antigen presentation and immune checkpoint expression. Targeting the RNF26-GRP78 axis represents a promising novel strategy for reversing HCC immune resistance and enhancing the efficacy of immunotherapy.

HSPA5 promotes YAP/TAZ stability independently of the Hippo pathway and induces proneural-to-mesenchymal transition in glioblastoma. (PubMed, Cell Death Dis)
Patients with high expression of HSPA5, YAP, and TAZ exhibited significantly poorer overall survival. Collectively, our findings suggested that HSPA5 promotes PMT through the stabilization of YAP/TAZ and identified HSPA5 as a promising therapeutic target for GBM patients.

Therapeutic optimization of LIPA targeting to induce endoplasmic reticulum stress and cell death in ovarian cancer. (PubMed, Oncogene)
We have previously shown that the tris-benzamide ERX-41 can induce ERS and cancer cell death in OCa by targeting LIPA. Immunohistochemical analysis of treated tumors demonstrated changes in expression of proliferative marker (ki67, decreased) and the ERS marker (GRP78, increased). These findings support the clinical advancement of ERX-208 for the treatment of patients with OCa.

circTNIK Promotes Carbon Nanotubes-Induced Lung Carcinogenesis via GRP78-Mediated Endoplasmic Reticulum Stress and Suppression of Type I Interferon Signaling. (PubMed, ACS Nano)
In clinical lung cancer tissue samples, circTNIK expression was positively correlated with GRP78 expression and negatively correlated with IFN-I signaling intensity, further supporting its oncogenic role in vivo. In summary, this study reveals that circTNIK plays a key role in CNTs-induced lung cancer development by regulating GRP78-mediated ER stress and IFN-I immunosuppression, providing a potential biomarker and therapeutic target for the early diagnosis and treatment of environmental-exposure-related lung cancer.

These data indicate that while BiP is essential for secretion of tick-borne orthoflavivirus NS1, it is not required for infectivity. The antiviral effect of YM-1 likely reflects inhibition of other chaperones or additional cellular targets.

Our findings highlight the beneficial role of TGP in preserving synaptic structural integrity and functionality and suggest a novel mechanism of synaptic dysfunction implicated in the pathophysiology of PSD. The results indicate that the regulation of the ERS signaling pathway by TGP presents therapeutic promise for the treatment of PSD.
