HOXC8 derived from cancer-associated fibroblasts regulates lung cancer cell malignant metastasis and ferroptosis by mediating the transcription of GCH1. (PubMed, Pathol Res Pract)
Our results indicate that CAFs-derived exosomes are a key source of HOXC8 in lung cancer cells. HOXC8 directly binds to the GCH1 promoter to activate its transcription, which in turn suppresses ferroptosis and promotes lung cancer progression. These findings contribute to the new intervention and treatment options for combating the malignant progression of lung cancer.

Mechanistically, HOXC8 transcriptionally activates TRIM22, leading to the ubiquitination and degradation of IκBα, which subsequently potentiates NF-κB signaling to drive stemness maintenance in CRC cells. These findings delineate a previously unrecognized HOXC8/TRIM22/NF-κB signaling axis that critically regulates tumor progression, offering a promising molecular target for therapeutic intervention in CRC.

To investigate the function of HOXC8 in NSCLC, we showed that knockdown of HOXC8 led to massive NSCLC cell death in a mechanism of pyroptosis because both YVAD, a caspase-1 (CASP1) inhibitor, and disulfiram, which prevents gasdermin D (GSDMD) pore formation, blocked cell death caused by HOXC8 depletion...Finally, we demonstrated that cholesterol-conjugated HOXC8 siRNA was able to slow down NSCLC tumorigenesis. This study suggests that HOXC8 participates NSCLC development by controlling CASP1 expression and pyroptosis.

N6-methyladenosine-modified circSLCO1B3 promotes intrahepatic cholangiocarcinoma progression via regulating HOXC8 and PD-L1. (PubMed, J Exp Clin Cancer Res)
Taken together, m6A-modified circSLCO1B3 was correlated with poor prognosis in ICC and promoted ICC progression not only by enhancing proliferation and metastasis via potentiating HOXC8 expression, but also by inducing immune evasion via antagonizing PD-L1 degradation. These results suggest that circSLCO1B3 is a potential prognostic marker and therapeutic target for ICC.
