    HGF/MET pathway is associated with poor efficacy of Immune checkpoint inhibitors (ICIs) in advanced-stage NSCLC (ESMO 2023)
    The contribution of HGF/MET to NSCLC response to ICIs was further investigated by culturing peripheral blood mononuclear cells (PBMC)stimulated with 10 μg/mL phytohemagglutinin (PHA) and/or pembrolizumab (20 nM). The cells were cultured with recombinant HGF or co-cultured with NSCLC patients-derived explants.in the presence of cMET inhibitors Crizotinib or SGX...Conclusions Our study found that high levels of HGF in the peripheral blood are associated with poor response to ICIs, disease progression, and shorter survival in patients with advanced NSCLC. Furthermore, we showed through in vitro experiments that inhibiting the HGF/MET signaling pathway could be a promising therapeutic approach to enhance the efficacy of ICIs in this patient population.

    ctDNA Dynamics, Prognostic Markers and Resistance Mechanisms in Tepotinib-Treated METex14 Skipping NSCLC in the VISION Trial (IASLC-WCLC 2023)
    In the largest on-treatment LBx biomarker dataset for a MET inhibitor in METex14 NSCLC, MR was associated with improved outcomes and TP53 mutation had negative prognostic significance. On-target secondary MET mutations and bypass pathway activation were potential resistance mechanisms.

    Association of Serum Hepatocyte Growth Factor Level with Systemic Inflammatory Biomarkers in Patients with Pancreatobiliary Cancer. (PubMed, Turk J Gastroenterol)
    The significant relationship between serum hepatocyte growth factor level and systemic inflammation markers in patients with pancreatobiliary cancer is shown for the first time in our study. This study, which showed a significant relationship between the presence of metastasis and serum hepatocyte growth factor level, suggests that serum hepatocyte growth factor level may be a prognostic biomarker in patients who are diagnosed with pancreatobiliary cancer.

    Cetuximab resistance induced by hepatocyte growth factor is overcome by MET inhibition in KRAS, NRAS, and BRAF wild-type colorectal cancers. (PubMed, J Cancer Res Clin Oncol)
    Patients with mCRC receiving cetuximab + FOLFIRI who presented with high plasma HGF levels had significantly worse PFS and OS. Cetuximab resistance induced by HGF was mediated by AKT and ERK activation and overcome by MET inhibition in vitro.

    Outcomes based on plasma biomarkers in METEOR, a randomized phase 3 trial of cabozantinib vs everolimus in advanced renal cell carcinoma. (PubMed, BMC Cancer)
    PFS and OS were improved with cabozantinib versus everolimus at high and low baseline levels of all biomarkers. Low baseline HGF was consistently identified as a prognostic biomarker for improved PFS or OS with cabozantinib or everolimus, supporting further prospective evaluation of the prognostic significance of HGF in advanced RCC.

    Bone marrow plasma level of decorin may be associated with improved treatment outcomes in a subset of multiple myeloma patients. (PubMed, J Formos Med Assoc)
    Our findings suggest that decorin may exert protective effects in this subset of MM patients.
