HHLA3 transcriptionally promotes GPX2 to enhance anti-oxidative capacity by attenuating KEAP1-mediated NRF2 ubiquitination in lung adenocarcinoma. (PubMed, Free Radic Biol Med)
Stabilized NRF2 transcriptionally activates GPX2 expression and enhances anti-oxidative capacity to promote LUAD progression. Our study not only uncovers a novel role of HHLA3 in LUAD but also suggests a series of therapeutic targets for precise diagnosis and targeted therapy of LUAD.

Identification and validation of prognostic genes and prognostic models associated with cutaneous melanoma and integrative stress response. (PubMed, Front Immunol)
RT-qPCR showed that prognostic genes were up-regulated in human melanoma cells compared to fibroblasts. The identification of ISR-featured prognostic genes and risk score stratification provide new insights into targeting SKCM and enhancing the efficacy of immunotherapy.

Spasmolytic Polypeptide-Expressing Metaplasia (SPEM): the hidden linchpin in gastric carcinogenesis beyond the correa cascade. (PubMed, Int J Surg)
This review synthesizes current understanding of SPEM's pathogenesis, molecular mechanisms, and clinical implications. Furthermore, it proposes integration of SPEM assessment into gastric cancer risk stratification protocols to enable earlier intervention strategies that interrupt carcinogenesis at its most vulnerable and potentially reversible stage.

Different forms of selenium improve laying performance and ovarian antioxidant capacity: regulating gut microbiota and ovarian metabolism. (PubMed, Poult Sci)
Compared with the sodium selenite (SeS), nano-selenium (SeN), selenomethionine (SeM), and selenium-enriched yeasts (SeY), as novel selenium sources, are increasingly popular in laying hen production...Meanwhile, the gut microbiota and differential metabolites of ovary were closely related to laying performance and antioxidant capacity of ovary. Therefore, 0.4 mg/kg SeN, SeM, and SeY showed an advantage on improving laying rate, ovary function, and gut microbiota in laying hens, and they could be potential additives using in laying hen production.

Targeting GPX2 to disrupt lipid homeostasis and enhance cisplatin sensitivity in diffuse gastric cancer. (PubMed, Cell Death Discov)
Notably, inhibiting GPX2 enhances the efficacy of cisplatin by sensitizing cancer cells to apoptosis. These insights identify GPX2 not only as a vital prognostic biomarker but also as a promising therapeutic target for overcoming cisplatin resistance in DGC, offering new avenues for treatment strategies.

GPX2 induces macrophage M2 polarization through the MIF signaling pathway to promote colorectal cancer progression. (PubMed, Int J Biol Macromol)
In vivo animal studies demonstrated accelerated tumor growth associated with GPX2 overexpression, correlating strongly with increased MIF signaling and M2 macrophage. Collectively, our results delineate a previously unrecognized GPX2-MIF-M2 macrophage axis that critically promotes CRC progression, positioning GPX2 as a potential therapeutic target for CRC intervention.

N6-Methyladenosine-Modified circNEK11 Promotes Hepatocellular Carcinoma Progression via the miR-1236-3p/GPX2 Axis. (PubMed, Cancer Sci)
Mechanistically, circNEK11 directly binds to miR-1236-3p to regulate GPX2 expression, thus promoting HCC progression and recurrence. Collectively, our findings suggest that circNEK11 is a potential biomarker and target for HCC diagnosis and treatment.

GPR44-Dependent Regulation of the Selenoproteome by eIF4a3 in Murine Acute Myeloid Leukemia-Initiating Stem Cells. (PubMed, Biol Trace Elem Res)
Given the role of selenium in redox balance, metabolism, and immune function, targeting selenium metabolism in GPR44-expressing AML may offer a novel therapeutic approach. Our findings reveal a previously unrecognized link between GPR44 signaling, selenium metabolism, and leukemia progression, warranting further studies to explore selenoprotein-targeting strategies for AML treatment.

N6-methyladenosine-modified GPX2 impacts cancer cell stemness and TKI resistance through regulating of redox metabolism. (PubMed, Cell Death Dis)
As a predominant oncogenic driver in non-small cell lung cancer (NSCLC), EGFR frequently undergoes amplification or mutation, with EGFR-tyrosine kinase inhibitors (EGFR-TKIs) like gefitinib and erlotinib constituting frontline therapy for advanced EGFR-mutant cases. In vivo, GPX2 deletion constrained glutathione metabolism and boosted the effectiveness of TKI in cell line-derived xenograft models. Collectively, these findings demonstrate that GPX2 serves as a positive regulator of both primary and acquired EGFR-TKI resistance and could be a promising therapeutic target for precise treatment of NSCLC.

Integrative single-cell RNA sequencing and bulk RNA sequencing reveals the characteristics of glutathione metabolism and protective role of GSTA4 gene in pancreatic cancer. (PubMed, Front Immunol)
The GSH metabolism-associated prognostic signature developed in this study effectively identifies high-risk patients with PC. As a prognostic protective factor, GSTA4 exhibits downregulated expression in PC tissues and suppresses tumor proliferation and migration, highlighting its potential as a therapeutic target.
