Transcriptomic analysis revealed no significant correlation between ovarian GLI1 fusion tumors and sclerosing stromal tumors. PTBP1 reduced KLF9 mRNA stabilization by binding to KLF9 mRNA, which promoted CRC cell stemness and DDP chemoresistance. PTBP1 reduced the stability of KLF9 mRNA, thereby relieving the transcriptional inhibition of FUBP1 by KLF9, leading to the development of stemness and DDP chemoresistance in CRC cells.

Significantly, the administration of the selective FUBP1 inhibitor, FUBP1-IN-1, is shown to effectively suppress CCDC183-AS1-induced PCa BM. These results shed light on the involvement of CCDC183-AS1 in enhancing osteoclastogenesis and the underlying mechanism in facilitating PCa BM, offering a potential avenue for therapeutic interventions.

Taken together, our findings reveal for the first time a new mechanism by which circACTN4 could promote oncogenesis and the development of BC by increasing the AcH4 of MYC target genes via TIP60. Therefore, circACTN4 could be a novel target for BC diagnosis and remedy.

- Ewing sarcoma tumors have a deficiency in POLθ expression and a corresponding loss of MMEJ activity - EWSR1 is a splicing factor that interacts with other splicing factors such as FUBP1 and KHSRP/FUBP2 to accurately splice the POLQ mRNA. - The EWS-FLI1 fusion oncoprotein, or loss of EWSR1, causes a splicing defect, leading to exon25 skipping of the POLQ pre-mRNA and loss of POLθ expression - The MMEJ deficiency of EwS cells results in cellular sensitivity to inhibitors of Homologous Recombination or Non-Homologous End-Joining - Exon 25 skipping of POLQ mRNA is a predictive biomarker for HR inhibitors in human cancers.

Furthermore, both COL12A1 and PLEC mRNA and protein levels were significantly associated with chemotherapy resistance. In summary, using MALDI-MSI, we have identified proteins, including COL12A1 and PLEC, associated with chemotherapy resistance to be further evaluated as HGSOC biomarkers and/or therapeutic targets.
