Mechanism of FOXP2 in the Hepatocellular Carcinoma Progression via Ferroptosis Through RBM15B-Mediated m6A Modification. (PubMed, Appl Biochem Biotechnol)
Overexpression of RBM15B or KDM4C attenuated ferroptosis and reversed the suppression of HCC cell growth induced by FOXP2 overexpression. In conclusion, FOXP2 may promote ferroptosis and inhibit cell proliferation in HCC by decreasing SLC7A11 expression via the RBM15B/KDM4C axis in an m6A-dependent manner.

Transcription factor FOXP2 attenuates the stemness of breast cancer cells by abolishing the transcription of Twist1. (PubMed, Cancer Genet)
The compensatory expression of Twist1 in FOXP2-overexpressing breast cancer cells counteracted FOXP2's inhibitory effects on the stemness of breast cancer cells. Together, the results showed that FOXP2 attenuated the stemness of breast cancer cells by abolishing the transcription of Twist1.

Notably, AKT activation enhanced FOXP2 expression, indicating a FOXP2/SOS1/AKT negative feedback loop. Collectively, our findings suggest that FOXP2 inhibits proliferation in KRAS-mutant CRC by suppressing SOS1-mediated PI3K/AKT signaling, and may serve as a prognostic biomarker and potential therapeutic target in KRAS-driven CRC.
