Compared with primary tumors, metastatic tumors demonstrated small but consistent genomic differences. These findings varied across cancer types and may reflect changes associated with the evolutionary transition from primary to metastatic disease.

Their consistent downregulation across different stages of HCC and their linkage to the tumor microenvironment underscore their potential role in HCC pathogenesis. These insights warrant further investigation and may contribute to the development of more effective diagnostic tools.

PROX1 expression is significantly associated with ERG expression and TMPRSS2-ERG fusion status in prostate cancer. These findings reinforce the role of PROX1 as an early marker and potential mediator of lineage plasticity. Targeting PROX1 or its regulatory pathways may offer a novel therapeutic strategy to mitigate plasticity-driven progression and treatment resistance in TMPRSS2-ERG fusion-positive prostate cancer.

The findings indicated that TPR and FGA exhibited good diagnostic performance, with AUCs of 0.946 (p = 0.002; 95% confidence interval [CI], 0.84-1.05) and 0.809 (p = 0.034; 95% CI, 0.65-0.97), respectively. Our results suggest that the TPR and FGA levels are potential predictors of post-EGFR-TKI treatment relapse.

PanScore is a validated deep learning model trained on genomic features that stratifies PDAC patients-particularly those with resectable disease- into biologically and prognostically distinct risk groups. It may inform personalized treatment decision-making beyond conventional staging.

FGA mRNA alone provides moderate diagnostic utility but substantially improves accuracy when combined with AFP, especially in low-AFP cases. This multi-biomarker approach holds promise for improving HCC detection and warrants further validation in larger cohorts.

In a mouse model of metastatic tumors, overexpression of FGA restricted the spread of tumor cells. In conclusion, FGA exhibits an inhibitory effect on tumor metastasis, providing new insights for the treatment of advanced HCC metastatic tumors.

The expression profiles and interactions of these proteins provide promising prognostic markers and therapeutic targets. These findings offer valuable insights into the post-translational modifications (PTMs) in human cancer, highlighting novel pharmacological and therapeutic opportunities.
