METTL14 facilitated the malignant growth of DTX-resistant cells and the polarization of M2 macrophages by modulating DKK4 expression. METTL14 targeting DKK4 influenced LC progression and DTX resistance, indicating that METTL14 could serve as a potential therapeutic target to overcome DTX resistance in clinical settings.

This study identified novel genes potentially associated with polyposis in patients lacking germline pathogenic variants in the APC and MUTYH genes. These findings support the use of next-generation sequencing for screening, expanding the scope of polyposis-related variants beyond these two genes.

This WNT-LEF1-dependent signature was also confirmed as activated in the hepatoblastoma tumor transcriptome. At the single-cell level, the WNT-LEF1 expression score was higher in tumor cells from both human hepatoblastoma samples and a hepatoblastoma patient-derived xenotransplant model; (4) This study uncovered a shared transcriptional activation of a LEF1-dependent embryonic program, which orchestrates the regulation of the Wnt signaling pathway in tumor cells from both hepatoblastoma and medulloblastoma.

First Transcriptome Analysis of Hepatoblastoma in Brazil: Unraveling the Pivotal Role of Noncoding RNAs and Metabolic Pathways. (PubMed, Biochem Genet)
In summary, our transcriptome analysis highlighted disrupted embryonic development as well as metabolic pathways, particularly those involving lipids, emphasizing the emerging role of ncRNAs as epigenetic regulators in hepatoblastomas. These findings provide insights into the complexity of the hepatoblastoma transcriptome and identify potential targets for future therapeutic interventions.

Colorectal cancer cells secreting DKK4 transform fibroblasts to promote tumour metastasis. (PubMed, Oncogene)
Reduced β-catenin activity by a chemical inhibitor MSAB promoted the CRC metastasis. Our findings demonstrate why reduced β-catenin activity is needed for CRC progression and provide a mechanism by which interactions between CRC cells and stromal cells affect disease promotion.

In addition, DKK4 low expression in the case of LARS high expression can be used as a poor prognostic factor in CRC patients. Thus, our findings suggest that DKK4 alone or in combination with LARS at diagnosis may be a useful prognostic factor for CRC.

O-GlcNAcylated LARP1 positively regulated by circCLNS1A facilitates hepatoblastoma progression through DKK4/β-catenin signalling. (PubMed, Clin Transl Med)
This study indicates that upregulated protein level of O-GlcNAcylated LARP1 mediated by circCLNS1A promotes the tumorigenesis and progression of HB through LARP1/DKK4/β-catenin axis. Hence, LARP1 and DKK4 are promising therapeutical target and diagnostic/prognostic plasma biomarker for HB.

Lastly, Mutational signature had difference in both groups, assuming that the early-onset group had been through different event attributing to mutational changes. Although these findings should be further validated by detailed functional experiments, our genomic and transcriptomic analysis provide additional insights into understanding of young CRC patients.

BRCA1 and BRCA2 mutations lead to Differential Wnt Signaling in Ovarian Cancer Cells (USCAP 2023)
Our findings indicate that specific loss of BRCA1 or BRCA2 show in ovarian cancer cells results in a differential response to Wnt signaling, which may be associated with difference in Wnt localization. This may contribute to the observed differences in the clinical outcome.

A new risk model for CSTA, FAM83A, and MYCT1 predicts poor prognosis and is related to immune infiltration in lung squamous cell carcinoma. (PubMed, Am J Transl Res)
The expression levels of CSTA, FAM83A, and MYCT1 are related to the diagnosis and prognosis of LUSC and may have potential as therapeutic targets in LUSC. A risk model and nomogram based on CSTA, FAM83A, and MYCT1 can predict the prognosis of LUSC.
