Enhanced Antitumor Immunity by ASP1570, a Novel Diacylglycerol Kinase Zeta Inhibitor, Offers a Potential Novel Immunotherapy for Treating Cancer. (PubMed, Mol Cancer Ther)
The antitumor efficacy of ASP1570 was cancelled by CD8+ T-cell depletion, indicating that its antitumor effect depends on CD8+ cytotoxic T-cell activation. Collectively, ASP1570 potentially improves antitumor efficacy in both anti-PD-1-therapy-resistant and anti-PD-1-therapy-responsive tumors by overcoming multiple immunosuppressive signals.

Finally, drug sensitivity data indicated that 10 chemotherapeutic drugs held promise as therapeutics for HCC patients with high-risk. In conclusion, multi-databases analysis showed that 5-CD8Gs and their signature could be an indicator to predict candidate drugs for HCC therapy.

Finally, the hub-regulated protease of diacylglycerol kinase zeta (Dgkz) was significantly identified and widely played various roles in glycerolipid metabolism, oxidative phosphorylation, and lipid kinase activation. Mitochondrial-regulated glycerolipid metabolism, especially the DGKZ protein, plays a key role in the metabolic reprogramming of immunotherapy-related myocarditis.

Taken together, these data suggest that BPA plays a role in colon carcinogenesis, and they provide insights into the molecular mechanisms of colon epithelial cell transformation by BPA. Increasing exposure to environmental toxins such as BPA can explain the increasing incidence of colorectal cancer.

DGKZ promotes TGFβ signaling pathway and metastasis in triple-negative breast cancer by suppressing lipid raft-dependent endocytosis of TGFβR2. (PubMed, Cell Death Dis)
Moreover, the caveolin/lipid raft-dependent endocytosis of TGFβR2 was regulated by the metabolite phosphatidic acid, which might alter TGFβR2 partitioning in lipid rafts and nonlipid rafts by affecting the fluidity of the plasma membrane. These findings suggested that DGKZ is a novel promoter of metastasis and that it could be a potential prognostic indicator in patients with triple-negative breast cancer.

Mechanisms of resistance to CLN-081 (TAS6417) in non-small cell lung cancer with EGFR exon 20 insertions (AACR 2022)
NSCLC with EGFR exon 20 insertions is resistance to conventional epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) such as osimertinib. Our preclinical study identified several mechanisms of resistance to CLN-081 including acquired mutations and inflammatory responses. These results will provide new insights into development of a novel strategy to overcome or prevent resistance to CLN-081.
