Mechanism of the DDX3X/NLRP3/GSDMD Signaling Axis in Pyroptosis and Inflammatory Response in Osteoarthritis Chondrocytes. (PubMed, J Biochem Mol Toxicol)
NLRP3 or GSDMD overexpression partly counteracted DDX3X knockdown-improved OA chondrocyte pyroptosis and inflammation. Inhibiting the DDX3X/NLRP3/GSDMD axis alleviated chondrocyte pyroptosis and inflammatory response in OA in vitro, thus ameliorating chondrocyte injury.

It presents a key diagnostic pitfall where secondary 11q aberrations in LBCL-IRF4-R can mimic HGBL-11q, revealing limitations of current genetic algorithms and advocating for expanded molecular diagnostic criteria.

Actinomycin D treatment combined with RT-qPCR was used to assess ILF3 mRNA stability...In vivo, DDX3X inhibitor, RK-33, markedly inhibited NB tumor growth and metastasis and enhanced M1 macrophage polarization, which was partially reversed by ILF3 overexpression. DDX3X promotes NB progression by stabilizing ILF3 mRNA, thereby facilitating M2 macrophage polarization.

OTUD4-ZMYND8-DDX3X Axis Drives Immunosuppressive Microenvironment in Spinal Metastases of Triple-Negative Breast Cancer. (PubMed, Neoplasia)
The resulting OTUD4-ZMYND8-DDX3X signaling axis drives canonical WNT/β-catenin signaling, upregulates CSF1 expression and promotes M2 polarization of macrophages, collectively fostering invasive behavior and establishing an immunosuppressive niche conducive to spinal metastasis. Collectively, these findings establish the OTUD4-ZMYND8-DDX3X axis as a pivotal regulator of spinal metastasis in TNBC and highlight its potential as a therapeutic target for inhibiting metastatic progression.
