DANCR (Differentiation Antagonizing Non-Protein Coding RNA)

This DANCR-mediated regulation promotes the interaction between ABL2 and the cytoskeletal regulator cortactin, which leads to activation of the SSH1-cofilin pathway and then facilitates the formation of lamellipodia, cytoskeletal reorganization and the metastatic ability of tumors. In summary, our findings delineate the DANCR/miR-125a-5p/ABL2/cofilin axis as a critical regulator of cytoskeletal dynamics in neuroblastoma metastasis, offering novel insights for the diagnosis and therapeutic targeting of high-risk neuroblastoma.

It is dynamically expressed across multiple different cell types in the developing embryo, transcriptionally activated by mitfa during early zebrafish development and it regulates genes involved in cell death. Our work suggests that cancer-critical lncRNAs such as DANCR, expressed from similar regions in vertebrate genomes, may control related genes and processes involved in both embryonic development and tumorigenesis across species.

lncRNAs are differently expressed and potentially play a regulatory role in the occurrence and development of NSCLC in older adults. Candidate lncRNA DANCR is highly expressed in older adults with NSCLC, which closely relates to clinical pathology and can contribute to progression-free survival and overall survival time prediction.

Though many of these potential associations are for uncharacterized transcripts, several identified lncRNAs (e.g., ERVH48-1 and CEBPA-DT) have been linked to pathways controlling cancer or placental growth, trophoblast differentiation, and gene expression. These associations warrant validation in future studies.

DANCR drives BCBM by functioning as a miR-758-3p sponge to upregulate PTGS2. Targeting the DANCR/miR-758-3p/PTGS2 axis represents a promising therapeutic approach.

Finally, the IC50 of 11 drugs had differences between high- and low-risk group, and only three of the drug's target genes (ERBB4, CASP8, and CD86) were differentially expressed. In conclusion, a prognostic model based on six feature lncRNAs (NFYC-AS1, OGFRP1, MIR4435-2HG, TDRKH-AS1, DANCR, and TMPO-AS1) was constructed by bioinformatics analysis, which might provide a new insight into the evaluation and treatment of LUAD.

Through bioinformatics analysis and experimental validation, we revealed that DANCR promotes ESCC progression by sponging miR-3193 and regulating its target gene DDIT3 expression. These findings highlight the critical role of DANCR in the development of ESCC and suggest its potential as a prognostic predictor and drug therapeutic target.

Bax and P21 protein levels were significantly upregulated following DNC treatment, whereas Bcl-2 and P53 protein levels were downregulated in DNC-treated breast cancer cells. In summary, dendrosomal nanocurcumin demonstrated potent anti-tumor effects against breast cancer cells, suggesting its potential as a therapeutic agent in breast cancer treatment.

This review aims to comprehensively explore the current knowledge regarding DANCR in CRC, including its molecular characteristics, expression patterns, and involvement in regulatory mechanisms, as well as its potential use as a diagnostic, prognostic, and therapeutic tool.
