SNX24 serves as a potential prognostic biomarker and a therapeutic target in uterine corpus endometrial carcinoma according to bioinformatics and experimentations. (PubMed, Transl Cancer Res)
Silencing of SNX24 significantly inhibited proliferation and invasion of endometrial carcinoma (EC) cells by downregulating the expression of CSNK1A1 and CSNK1E. Taken together, our data demonstrated the prognostic significance and tumor-promoting role of SNX24 in UCEC.

The role of MCPIP1 in key elements of the epithelial-mesenchymal transition signaling axis in clear cell renal cell carcinoma (PubMed, Postepy Biochem)
The obtained results indicate that the lack of MCPIP1 RNase activity activates genes and processes associated with the migratory activity of cancer cells. In summary, the results obtained in this doctoral thesis indicated that MCPIP1 may regulate the progression of clear cell renal cancer at various levels of proangiogenic and prometastatic factors, as well as by influencing the EMT process.

Druggable genome CRISPR screening identifies the KEAP1/NRF2 axis as a mediator of PD-L1 expression. (PubMed, Commun Biol)
Supporting this, pharmacological NRF2 activation resulted in significant antitumor immunity with increased cytotoxic effector T cell infiltration and reduced exhausted T cells, resulting in smaller xenografted tumors. These findings establish the KEAP1/NRF2 axis as a novel and potentially druggable mechanism of IFNγ-meditated PD-L1 expression in cancer cells.

Lenalidomide can selectively suppress the del(5q)-mutant clone by promoting proteasomal degradation of casein kinase 1A1 and inducing mutant stem cell failure...Accurate assessment of the TP53 allelic state is crucial for distinguishing MDS-del(5q) from TP53-mutant MDS, a myeloid malignancy characterized by TP53 multihit state and very aggressive clinical course. Genomic profiling is also critical for therapeutic decision making in patients with MDS-del(5q), particularly for assessing a patient's eligibility for allogeneic transplantation, which remains the only curative treatment.

Casein kinase 1α essentially regulates thrombopoiesis by driving megakaryocyte maturation and cytoskeleton organization. (PubMed, Blood)
Our findings could be translated into a human approach, since a CRISPR/Cas9-mediated genetic deletion of CSNK1A1 in MKs derived from human CD34+ progenitor cells resulted in a substantial defect in human MK maturation and platelet production. The present observations elucidated CK1α as an important signaling molecule in MK cytoskeletal dynamics and polarization, proplatelet formation and polyploidization, thus highlighting the crucial role of CK1α in platelet biogenesis.

Resistant del(5q) MDS-L cells, as well as primary MDS marrow cells, are also sensitive to targeting of IGF-1R-related dependencies in del(5q) MDS, which include the Abl and MAPK signaling pathways. This work thus provides potential new therapeutic avenues for lenalidomide-resistant del(5q) MDS.

Inhibition of CK1α using short hairpin RNA (shRNA) and treatment of a CK1α inhibitor or an mTOR inhibitor, TORKinib, resulted in decreased β-TrCP interaction with LT, increased LT expression, and enhanced MCPyV replication...Therefore, cellular kinase pathways are indispensable for governing MCPyV polyomavirus infection and life cycle in coordinating with the immunosuppression environment at disease onset. Understanding the regulation mechanisms of MCPyV replication by viral and cellular factors will guide proper prevention strategies with targeted inhibitors for MCPyV-associated Merkel cell carcinoma (MCC) patients, who currently lack therapies.

Downregulation of Myc protein expression correlated with efficient elimination of blasts in A51 treatment. The "myc signature" closely resembled the transcriptional profile of del(5q) MDS patients with TP53 mutation.

Axin phosphorylation in condensates counteracts tankyrase-mediated degradation. (PubMed, J Cell Sci)
Importantly, the phosphorylation took place within the tankyrase-binding site, hindered axin-tankyrase interaction electrostatically and/or sterically, and counteracted tankyrase-mediated degradation of axin. Thus, the presented data propose a novel mechanism regulating axin stability, with implications for Wnt signaling, cancer therapy and self-organization of biomolecular condensates.

Oncogenic K-Ras suppresses global miRNA function. (PubMed, Mol Cell)
Hypo-phosphorylated Ago2 increased binding to mRNAs while reducing its activity to repress miRNA targets. Our findings connect a potent regulatory mechanism of global miRNA activity to K-Ras in a pathophysiological context and provide a mechanistic link between oncogenic K-Ras and the post-transcriptional upregulation of miRNA targets.
