We found that these GRP78 inhibitors regressed pediatric and adult brain tumor neurospheres and eliminated ROR1, Cripto, and PD-L1 expression, leading to tumor cell apoptosis. Cell surface GRP78 inhibition with systemic dosing of GRP78 inhibitors, in preclinical orthotopic brain tumor mouse models, demonstrated significant increases in overall survival and durable regressions in 40-70% of treated mice compared to control mice.

Transcriptomic analyses revealed a unique CRIPTO/MYC co-activation signature associated with PSA progression in a human PCa cohort. Taken together, our data highlights a role for CRIPTO in tumor invasiveness and progression, which carries implications for biomarkers and targeted therapies.
Our discriminatory MoAbs provide new reagents to help clarify current CR1/CR3 protein expression vagaries in the Cripto field of study, challenging established CR1 conventions. In addition, our data validate CR3 involvement in human carcinogenesis and cell signaling pathways, with potential clinical relevance in determining cancer patient prognosis and disease severity.
