Knockdown of Claudin-8 (CLDN8) Indicates a Link Between Breast Cancer Cell Sensitivity to Chemotherapeutics and Reveals a Potential Use of CLDN8 as a Molecular Diagnostic and Target for Therapy. (PubMed, Int J Mol Sci)
In conclusion, our findings identify CLDN8 as an important prognostic factor in breast cancer and as a novel predictor of treatment response. These results underscore the potential utility of CLDN8 status in guiding personalized therapy and highlight CLDN8 as a candidate target for overcoming treatment resistance in breast cancer.

Deep learning assisted identification of SCUBE2 and SLC16 A5 combination in RNA-sequencing data as a novel specific potential diagnostic biomarker in prostate cancer. (PubMed, Med Biol Eng Comput)
In summary, this study identifies SCUBE2 as a novel potential diagnostic biomarker for prostate cancer and supports the use of AI-driven gene discovery in identifying key players in tumor biology. The combination of SCUBE2 with SLC16A5 not only enhances diagnostic precision but also opens new avenues for functional and clinical validation, ultimately contributing to the development of more accurate, multi-gene diagnostic panels for PCa.

IL-4, IL-13, TNF-α and IFN-γ Downregulate CLDN8 Expression Through Activating JAK Signaling Pathway in HaCaT Cells. (PubMed, Clin Cosmet Investig Dermatol)
CLDN8 mRNA expression is decreased in AD lesions and MC903-induced AD-like mouse model. Downregulation of CLDN8 mRNA expression is alleviated after dupilumab or crisaborole treatment...Skin tissues might not express CLDN8 protein. AD-related cytokines including IL-4, IL-13, TNF-α and IFN-γ could downregulate CLDN8 mRNA expression through activating JAK.

The immunological landscape of CCL26High invasive oral squamous cell carcinoma. (PubMed, Front Cell Dev Biol)
However, CCL26 did not affect T/B/NK lymphocytes in peripheral blood of OSCC patients. CCL26 could regulate Immune balance and promote invasiveness of OSCC, which gave a new insight into a potential immunotherapy strategy.

Magnetic Resonance Imaging-Based Radiogenomic Analysis Reveals Genomic Determinants for Nanoparticle Delivery into Tumors. (PubMed, ACS Nano)
By performing Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses of genes and pathways, we identified a variety of genes affecting vascular permeability, such as Cldn1, Dlg2, Bves, Prkag3, Cldn10, and Cldn8, which are related to tight junctions and control the permeability of blood vessels in tumors. The method presented here provides an MRI-supported approach to increase the breadth of data collected from genetic screens, reveals genetic evidence of the presence of NPs in tumors and lays a foundation for clinical patient stratification and personalized treatment.

Bioinformatics analysis of colorectal cancer transcriptomic data reveals novel prognostic signature and potential biomarker genes. (PubMed, Scand J Gastroenterol)
Increased expression of REG1A, MMP3, FOXQ1 and CEMIP genes and decreased expression of AQP8, CA1, CLDN8, PYY, CA4, CEACAM7 and SLC30A10 genes were observed. This approach revealed a CRC-specific molecular profile and may provide some guidance for further investigation of potential biomarkers for diagnosis and prognosis prediction of CRC patients.

CSER: a gene regulatory network construction method based on causal strength and ensemble regression. (PubMed, Front Genet)
Here, using real gene expression data, we applied CSER to construct a colorectal cancer GRN and successfully identified several key regulatory genes closely related to colorectal cancer (CRC), including ADAMDEC1, CLDN8, and GNA11. Importantly, by integrating immune cell and microbial data, we revealed the complex interactions between the CRC gene regulatory network and the tumor microenvironment, providing additional new biomarkers and therapeutic targets for the early diagnosis and prognosis of CRC.

Significance and Possible Biological Mechanism for CLDN8 Downregulation in Kidney Renal Clear Cell Carcinoma Tissues. (PubMed, World J Oncol)
CLDN8 is downregulated in KIRC and is considered a potential tumor suppressor. CLDN8 deficiency may promote the initiation and progression of KIRC, potentially in conjunction with metabolic dysfunction.

Heterogenous Gene Expression of Bicellular and Tricellular Tight Junction-Sealing Components in the Human Intestinal Tract. (PubMed, Biol Pharm Bull)
Considering their segment specificity, claudin-8, claudin-15, and angulin-2 appear to be targets for the development of permeation enhancers in the rectum, small intestine, and large intestine, respectively. These data on heterogenous expression profiles of intestinal TJ components will be useful for the development of safe and efficient intestinal permeation enhancers.

High expressions of CLDN8, CTNNA3, MYL2 and MYLPF genes in patients with oral lichen planus have a good clinical response to topical dexamethasone, while patients with high expression genes of inflammation pathway such as TNF, IL-17, NF-κB and cortisol synthesis and secretion received poor effect.

In vitro model of human mammary gland microbial colonization (MAGIC) demonstrates distinctive cytokine response to imbalanced human milk microbiota. (PubMed, Microbiol Spectr)
The MAGIC cell model also offers new opportunities for research in other areas of MG physiology, such as the effects of bioactive milk components on microbial colonization of the MG, mastitis prevention, and studies of probiotic development. Since resident MG bacteria may be an important factor in breast cancer development, the MAGIC in vitro tool also offers new opportunities for cancer research.
